Background Previous observational epidemiological studies reported an association between cathepsins and cancer, however, a causal relationship is uncertain. This study evaluated the cancer using Mendelian randomization (MR) analysis. Methods We used publicly available genome-wide (GWAS) data for bidirectional MR Inverse variance weighting (IVW) was as primary method of Results After correction False Discovery Rate (FDR), two were found to be significantly associated with risk: cathepsin H (CTSH) levels increased risk lung (OR = 1.070, 95% CI 1.027–1.114, P 0.001, FDR 0.009), CTSH decreased basal cell carcinoma 0.947, 0.919–0.975, 0.0002, 0.002). In addition, there no statistically significant effect 20 cancers on nine cathepsins. Some unadjusted low P-value phenotypes are worth mentioning, including positive correlation O (CTSO) breast 1.012, 1.001–1.025, 0.041), S (CTSS) pharyngeal 1.017, 1.001–1.034, 0.043), CTSS endometrial 1.055, 1.012–1.101, 0.012); negative Z ovarian (CTSZ) 0.970, 0.949–0.991, 0.006), prostate 0.902–0.944, 0.028), E (CTSE) pancreatic 0.963, 0.938–0.990, 0.006). Conclusion Our analyses showed may help provide new insights further mechanistic clinical cathepsin-mediated cancer.

Abstract Objective To investigate the genetic underpinnings of association between type 2 diabetes (T2D), glycemic indicators such as fasting glucose (FG), insulin (FI), and glycated hemoglobin (GH), venous thromboembolism (VTE), encompassing deep vein thrombosis (DVT) pulmonary embolism (PE), thereby contributing novel insights to scholarly discourse within this domain. Methods Genome-wide study (GWAS) summary data pertaining exposures (T2D, FG, FI, GH) outcomes (VTE, DVT, PE) were acquired from IEU Open GWAS database, participants European descent, including both male female individuals. Two-sample Mendelian randomization (MR) analyses conducted utilizing TwoSampleMR MRPRESSO packages R programming environment. The primary analytical approach employed was random-effects inverse variance weighted (IVW) method. Heterogeneity assessed via Cochran’s Q statistic for MR-IVW Rucker’s MR-Egger. Horizontal pleiotropy evaluated using intercept test MR Egger residual sum outlier (MR-PRESSO) analysis, with latter also detection. Additionally, a “Leave one out” analysis ascertain influence individual single nucleotide polymorphisms (SNPs) on results. Results IVW revealed negative causal T2D) VTE ( P = 0.008, Odds Ratio [OR] 95% confidence interval [CI] 0.896 [0.827–0.972]), well FG 0.002, OR CI 0.655 [0.503–0.853]), GH 0.010, 0.604 [0.412–0.884]), DVT 0.413 [0.235–0.725]). Abstract Background Association between glucose and inflammatory bowel disease (IBD) was found in previous observational studies cohort studies. However, it is not clear whether these associations reflect causality. Thus, this study investigated there such a causal relation elevated IBD, Crohn’s (CD) ulcerative colitis (UC). Methods We performed two-sample Mendelian Randomization (MR) with the independent genetic instruments identified from largest available genome-wide association (GWAS) for IBD (5,673 cases; 213,119 controls) its main subtypes, CD UC. Summarized data which included 200,622 cases glycemic traits including HbA1c type 2 diabetes(T2DM) were obtained different GWAS Primary secondary analyses conducted by preferentially using radial inverse-variance weighted (IVW) approach. A number of other meta-analysis approach sensitivity carried out to assess robustness results. Results did find effect genetically predicted on as whole (OR 0.858; 95% CI 0.649–1.135; P = 0.286). In subtype also suggestively associated 0.22; 0.04-1.00; 0.05) 0.940; 0.628–1.407; 0.762). direction, subtypes related traits. Conclusions This MR providing any evidence relationship well it’s UC CD. Regarding no could be found. Future robust are needed confirm conclusion.
