ABSTRACT Skeletal insufficiency affects all individuals with Down syndrome (DS) or trisomy 21 and may alter bone strength throughout development due to a reduced period of formation early attainment peak mass compared those in typically developing individuals. Appendicular skeletal deficits also appear males before females DS. In femurs male Ts65Dn DS model mice, cortical were pronounced development, but trabecular Dyrk1a overexpression transitory until postnatal day (P) 30, when there persistent was trending toward overexpression. Correction DS-related by purported DYRK1A inhibitor through genetic means beginning at P21 not effective P30, germline normalization improved structure P36. Trabecular female mice evident P30 subsided P36, typifying periodic developmental normalizations that progressed more prominent deficiencies. Sex-dependent differences delayed impact trisomic are important find temporally specific treatment periods for other phenotypes associated 21.
