Abstract Originating from, but independent of, linear chromosomes, extrachromosomal DNA (ecDNA) exists in a more active state of transcription and autonomous replication. It plays crucial role the development malignancies therapy resistance. Since its discovery eukaryotic cells than half century ago, biological characteristics functions ecDNA have remained unclear due to limitations detection methods. However, recent advancements research tools transformed research. is believed that exhibits greater activity abnormal amplification oncogenes, thereby driving cancer progression through their overexpression. Notably, compared DNA, can also function as genomic element with regulatory roles, including both trans‐ cis‐acting functions. Its critical roles tumorigenesis, evolution, progression, drug resistance malignant tumors are increasingly recognized. This review provides comprehensive summary evolutionary context highlights significant progress understanding potential applications therapeutic target tumors.
