Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by progressive polyarthritis that leads to cartilage and bone damage. Pre-clinical RA prolonged state before clinical develop, in which autoantibodies (antibodies against citrullinated proteins, rheumatoid factors) can be present due the breakdown of immunologic self-tolerance. As early treatment initiation onset may achieve sustained remission, optimize outcomes, even prevent progression, pre-clinical stage showing prospect window opportunity for treatment. Growing evidence has shown role gut microbiota inducing systemic inflammation via multiple mechanisms, involve molecular mimicry, impaired intestinal barrier function, microbiota-derived metabolites mediated immune regulation, modulation microbiota's effect on cells, epithelial cells autophagy, interaction between microbiome human leukocyte antigen alleles as well microRNAs. Since alterations have been reported, potential therapies modifying RA, including natural products, antibiotic therapy, fecal transplantation, probiotics, microRNAs vitamin D supplementation, autophagy inducer-based treatment, prebiotics, diet, holds great promise successful prevention altering ongoing inflammation. In this review, we summarized current studies include pathogenesis progression promising therapeutic strategies provide novel ideas management possibly preventing progression.

Abstract MicroRNAs (miRNAs), a class of endogenous single-stranded short noncoding RNAs, have emerged as vital epigenetic regulators both pathological and physiological processes in animals. They direct fundamental cellular pathways by fine-tuning the expression multiple genes at posttranscriptional level. Growing evidence suggests that miRNAs are implicated onset development rheumatoid arthritis (RA). RA is chronic inflammatory disease mainly affects synovial joints. Ulcerative colitis (UC), an inflammatory bowel disease (IBD), may increase the risk of colorectal cancer (CRC) by activating chronic proinflammatory pathways. The goal this study was to find serum prediction biomarkers in UC CRC development combining low-density miRNA microarray and biocomputational approaches. expression profiles were compared microarray, finding five upregulated miRNAs specific progression (hsa-let-7d-5p, hsa-miR-16-5p, hsa-miR-145-5p, hsa-miR-223-5p, hsa-miR-331-3p). circRNA/miRNA/mRNA competitive endogenous RNA (ceRNA) network analysis showed that candidate connected well-known colitis-associated ACVR2A, SOCS1, IGF2BP1, FAM126A, CCDC85C mRNAs, circ-SHPRH circRNA. SST SCARA5 genes regulated hsa-let-7d-5p, hsa-miR-331-3p linked a poor survival prognosis patient dataset from Cancer Genome Atlas (TCGA). Lastly, our mRNA candidates validated comparing their differentially expressed mRNAs tissue databases. A high level parallel reduction SOCS1 found serum. We propose possibly hsa-let-7d-5p as novel for predicting CRC. More clinical sample is required further validation.

Rheumatoid arthritis (RA) is an autoimmune disease that causes inflammation, pain, and ultimately, bone erosion of the joints. The this are multifactorial, including genetic factors, such as presence human leukocyte antigen (HLA)-DRB1*04 variant, alterations in microbiota, or immune factors increased cytotoxic T lymphocytes (CTLs), neutrophils, elevated M1 macrophages which, taken together, produce high levels pro-inflammatory cytokines. In review, we focused on function exerted by osteoclasts osteoblasts other means release exosomal microRNAs (miRNAs). Based a thorough revision, classified these molecules into three categories according to their function: osteoclast inhibitors (miR-23a, miR-29b, miR-214), osteoblast (miR-22-3p, miR-26a, miR-27a, miR-29a, miR-125b, miR-146a), enhancers (miR-20a, miR-34a, miR-96, miR-106a, miR-142, miR-199a, miR-324, miR-486b). Finally, analyzed potential therapeutic targets miRNAs, use antagomiRs, blockmiRs, agomiRs competitive endogenous RNAs (ceRNAs), which already being tested murine ex vivo models RA.
