Abstract Long non-coding ribonucleic acids (RNAs) (lncRNAs) are key players in tumorigenesis and immune responses. The nature of their cell type-specific gene expression other functional evidence support the idea that lncRNAs have distinct cellular functions tumor microenvironment (TIME). To date, majority lncRNA studies heavily relied on bulk RNA-sequencing data which various types contribute to an averaged signal, limiting discovery functions. Single-cell (scRNA-seq) is a potential solution for tackling this limitation despite lack annotations low abundance yet lncRNAs. Hence, updated further understanding will be necessary characterizing genes TIME. In review, we discuss specifically expressed cells, summarize regulatory at type level highlight how scRNA-seq approach can help study TIME

Abstract Cervical cancer (CC) poses a significant threat to women's health and lives worldwide. Emerging evidence indicates that long noncoding RNA LINC01871 is closely associated with immune regulation in CC patients. However, its specific role underlying mechanisms remain poorly understood. In this study, we examined the expression levels subcellular localization of cell lines. Functional assays, including transwell migration invasion assays as well macrophage phagocytosis were conducted estimate invasiveness, migration, response. Western blotting was performed assess protein markers epithelial–mesenchymal transition (EMT), immunity, MAPK signaling. A luciferase reporter assay used confirm interactions between LINC01871, miR‐873‐3p, MAP3K2. Additionally, xenograft mouse model employed investigate vivo effects on progression. Our results revealed highly expressed predominantly localized cytoplasm knockdown significantly suppressed invasion, EMT, escape vitro reduced tumor growth vivo. Mechanistically, found interact leading upregulation MAP3K2 subsequent activation Furthermore, overexpression rescued inhibitory silencing malignant behaviors cells. conclusion, facilitates metastasis by driving EMT modulating response via miR‐873‐3p/MAP3K2/MAPK signaling pathway.
