Abstract Purpose To compare the course of retinal ganglion cell (RGC) loss following optic nerve crush (ONC) in male and female mice to assess neuroprotective potential intravitreally administered oestrogen receptor alpha (ERα) or beta (ERβ) agonists. Methods Optic was performed on left C57BL/6 mice. RGC evaluated at 3–21 days post‐lesion. ERα ERβ agonists (PPT DPN, respectively) were injected either immediately after ONC with retinas analysed 5 post‐ONC again 9 days, 21 Intact served as controls. The total population Brn3a + RGCs quantified flat mounts. expression ERβ, immunodetection cross sections, percentage expressing each determined Results Retinal both sexes followed two distinct linear phases: A rapid phase up post‐lesion, by a slower phase. However, onset earlier males than females. Both expressed all layers majority RGCs. While agonism did not confer protection, during second axotomy‐induced loss. Conclusions Preclinical results may be influenced pooling data from effects loss, likely linked neuroinflammation, suggest therapeutic applications for chronic conditions such glaucoma.
