Chronic inflammation is an important component of many diseases, including autoimmune intracellular infections, dysbiosis and degenerative diseases. An element this state the mainly positive feedback between inflammatory cytokines, reactive oxygen species (ROS), nitric oxide (NO), increased calcium, hypoxia-inducible factor 1-alpha (HIF-1α) stabilisation mitochondrial oxidative stress, which, under normal conditions, enhance response against pathogens. Autophagy nuclear erythroid 2-related 2 (Nrf2)-mediated antioxidant are negatively coupled with above-mentioned elements to maintain defence at a level appropriate severity infection. The current review first attempt build multidimensional model cellular self-regulation chronic inflammation. It describes feedbacks involved in explains possible pathways by which becomes chronic. multiplicity suggests that symptomatic treatment should focus on inhibiting multiple effectively suppress all dysregulated inflammation, calcium mito-stress other metabolic disturbances.

The intestinal barrier function is a critical defense mechanism in the human body, serving as both primary target and initiating organ cases of sepsis. Preserving integrity this essential for preventing complications diseases, including sepsis mortality. Despite importance, impact resveratrol on remains unclear. Thus, study aims to explore potential beneficial effects maintaining function. Fifteen male Sprague Dawley rats, weighing between 180 g 220 g, were randomly assigned one three groups: control group (Con), lipopolysaccharide (LPS) group, (RSV) group. received an intravenous administration at dosage 8 mg/kg, 10 min prior treatment. Each comprised five rats. Various techniques enzyme-linked immunosorbent assay (ELISA), hematoxylin eosin staining (HE), periodic acid Schiff (PAS) staining, transmission electron microscopy (TEM), Western blot analysis (WB), quantitative real-time polymerase chain reaction (qRT-PCR) utilized assess differences inflammatory cytokine expression, histopathological changes, apoptosis, tight junction (TJ) protein, TLR4/MyD88/NF-кB signaling pathways. Resveratrol exhibited anti-inflammatory by decreasing levels interleukin (IL)-1β, interleukin(IL)-6, tumor necrosis factor (TNF)-α, while increasing (IL)-10. Additionally, rats treated with resveratrol, there was reduction expression apoptosis-associated proteins Bax Caspase-3. also significantly increased (TJ), decreased fatty binding protein (I-FABP) D-lactic (D-LA). Furthermore, related pathways TLR4, MyD88, NF-κB decreased. has been shown reduce apoptotic proteins, enhance inhibit response mediated TLR4/MyD88/NF-κB pathway, thereby alleviating LPS-induced septic injury.

Aging is a very complex process, and it has been linked with Sirtuins. Sirtuin enzymes are family of deacetylases that related to caloric restriction aging by modulating energy metabolism, genomic stability, stress resistance. Up now, seven sirtuins have recognized. This narrative review aimed analyze the literature produced between January 2005 March 2025 evaluate role in chronic kidney disease and, as heart diseases strictly interrelated, explore their cardio-renal cross-talk. A reciprocal relationship CKD seems exist since may contribute premature biological different organ systems. SIRTs involved pathophysiology renal diseases; activation can delay progression several diseases. Notably, an increasing number studies CVDs. affect production mitochondrial reactive oxygen species (ROS) function. The imbalance SIRT levels increase vulnerability pathophysiological mechanisms HFpEF (heart failure preserved ejection fraction) through signaling pathways. Fibrosis linkage mechanism development Current on sirtuins, resveratrol, cardiorenal highlight potential therapeutic benefits regulating blood pressure, function, lipid profiles, inflammation, making them promising area investigation for improving cardiovascular health outcomes. However, significant gaps remain. limited availability highly selective potent sirtuin modulators hampers clinical translation, most existing compounds exhibit poor bioavailability suboptimal pharmacokinetic properties.
