Rheumatoid Arthritis (RA) is a persistent autoimmune inflammatory disorder that arises from the intricate interaction between genetic predisposition and environmental influences. The progression of RA can be delineated into four distinct phases: initially, influence risk factors; followed by emergence systemic autoimmunity; subsequently, an asymptomatic phase; ultimately, manifestation clinical arthritis. Recently, role mucosal immunity in has gained significant attention research. Evidence published studies suggests not only influences onset but also plays crucial its progression. Scholars have begun to unravel links barriers gastrointestinal tract, respiratory system, oral cavity. Specifically, shifts microbiota, dysfunction barriers, abnormal activation immune tissues are all implicated pathogenesis RA.Despite this growing body knowledge, comprehensive review therapeutic implications yet conducted. ABSTRACT Rheumatoid arthritis (RA) is a severe inflammatory autoimmune disease with metabolic changes. RA patients have abnormalities in glycolysis, amino acid metabolism, choline and fatty synthesis. The differential metabolites individuals of animal models were explored to find the potential biomarkers for risk prediction, diagnosis, prognosis perspective metabolism. Moreover, we discussed changes related after treatment anti‐rheumatic drugs, Traditional Chinese Medicine (TCM) explore promising metabolites. In addition, immunological mechanism TCM from metabolism was also clarified. For perspectives research application beneficial clinic, relevant technologies focuses future studies field been proposed accordingly.

Rheumatoid arthritis (RA) is a chronic autoimmune disease that causes synovial joint inflammation as well bone destruction and erosion, typically characterized by pain, swelling, stiffness, with complications persistent pain after remission posing significant health burden for RA patients. The etiology of has not yet been fully elucidated, but large number studies have shown the initiation in closely related to T-cell activation, production variety pro-inflammatory cytokines, macrophage M1/M2 imbalance, homeostatic imbalance intestinal flora, fibroblast-like synoviocytes (FLSs) tissue macrophages (STMs) lumen joints exhibit an aggressive phenotype.
