Listeria monocytogenes is not only a pathogen causing serious food-borne disease in humans but can also occur as saprophyte the natural environment. This study aimed to evaluate occurrence of L. monocytogenes obtained from environment Czech Republic (hereinafter Czechia) and Slovakia 2016–2018 compare clonal relationship strains circulating with originating food chain humans. Altogether, 217 samples mud, surface water, vegetation soil were collected 61 locations. Samples processed according modified EN ISO 11290-1 standard. The isolates characterised using serotyping, macrorestriction analysis, followed by pulsed-field gel electrophoresis, whole genome sequencing (WGS), antimicrobial susceptibility testing. L. detected 8.8% examined isolated 15 locations, mainly mud banks water sources. 25 19 positive samples. Serotypes 1/2a, 4b, 1/2b among strains. Twenty combined AscI/ApaI pulsotypes analysis. 12 sequence types (STs) Multi-locus typing (MLST) WGS, ST451 being most frequent. core MLST analysis revealed heterogeneous population environmental No phenotype resistance was Screening antimicrobial-resistance genes platform ResFinder fosX 24 blaTEM-116 one isolate. various environments within wide altitude range during different seasons year may highlight this bacterium's remarkable adaptability exceptional tolerance external factors. Serotype distribution Czechia seems reflect human more than chain.

The presence of Listeria monocytogenes in the dairy environment remains a food safety challenge. source microbial contamination may include employees and their personal protective equipment (PPE). This study investigated effectiveness cleaning protocols (i.e., detergents mechanical action) three chemical sanitizers commonly employed at facilities against innocua on different types gloves, aprons, boots. Coupons made PPE material were inoculated with two-strain cocktail L. suspended either Phosphate Buffered Saline (PBS) or skim milk to determine potential effect organic matter. In general, peroxyacetic acid (0.20% (v/v)) was more effective reducing counts aprons gloves compared chlorine quaternary ammonium 200 ppm. Depending type sanitizer, reductions ranged from 1.95 - 4.72 1.52 4.60 log CFU/in2 respectively. comparison, achieved 0.93 2.32 reduction boot soles, no significant differences observed among sanitizers. PVC (vinyl) lower than nitrile latex gloves. Sanitizers less boots wider deeper lugs those shallow lugs. matter significantly reduced antimicrobial efficacy all (<1 reduction). However, inclusion without action ≥3 PPE. highlights importance scrubbing as an essential step reduce

Abstract Background Listeriosis is a global health threat to both animals and humans, especially in developing countries. This study was designed isolate Listeria monocytogenes from faeces; environmental samples; cow, sheep goat milk, as well human stool, its molecular characteristics antibiotic sensitivity the New Valley Beheira Governorates, Egypt. The isolation identification of L. were carried out using traditional culture biochemical methods, followed by antibiography, genus confirmation some isolates detection sequencing InlB genes via PCR. Results Out 2097 examined samples, prevalence 13.4% animals; 9.2%, 2.4%, 25.4%, 4%, 42.4%, 6.4% cattle faeces, respectively. However, 8.3% samples. Both animal showed 100% resistance trimethoprim-sulfamethoxazole, highest flumequine (100%), amikacin (99.2%), gentamicin (97.6%), levofloxacin (94.6%). Multidrug (MDR) detected 86.9% tested isolates. 16 S rRNA inlB randomly selected Phylogenetic analysis three based on gene identity between faecal, milk stool Conclusions Faeces are major sources listeriosis, high degree genetic similarity suggests possibility zoonotic circulation. MDR samples could negatively impact success prevention treatments for diseases, thereby imposing serious risks public health.

The close genetic resemblance between Listeria monocytogenes and innocua, combined with their presence in similar environments, poses challenges for species-specific detection food products. Ensuring safety through microbiological standards necessitates reliable of pathogens like L. innocua throughout the chain using appropriate analytical techniques. This study aims to develop, identify, validate a SYBR Green qPCR-based marker designed detect innocua. By performing comparative analysis complete genome sequences (ATCC 12392) (CFSAN044836), unique gene region encoding hypothetical protein an LPXTG cell wall anchor domain (GCF_003031895.1) leucine-rich repeats (GCF_009648575.1) was identified. Primers targeting these specific were validated effectiveness detecting monocytogenes/L. both conventional PCR qPCR These primers exhibited high sensitivity specificity amplifying among different species. further confirmed standard curve three templates: cloned DNA (as positive control), genomic DNA, bacterial suspension. Additionally, rigorously tested accuracy directly targeted strains live enoki mushroom samples. direct method offers significant advantages rapid precise potentially enhancing efficiency diagnostic monitoring processes within vegetable distribution systems.

With the rapid implementation of high-pressure processing in many sectors food industry, considerations associated with pressure-stressed microorganisms are emerging. Nisin was utilized this study for controlling proliferation Listeria monocytogenes and L. innocua inoculated on cold-smoked trout during a 4-week refrigerated shelf-life trial. Wild-type phenotypes were compared study. The prepared by treating surrogate strain pathogen mixture at 103.4 MPa (15K PSI) 20 min. multiplied extensively trial counts increased (p < 0.05) from 3.68 ± 0.1 log CFU/g first week to 6.03 CFU/g. Both illustrated similar ≥ multiplication trends. Unlike samples subjected water treatment, nisin effective keeping microbial lower controls, particularly earlier Our illustrates that selected microorganism has comparable sensitivity relative thus could be used interchangeably future public health microbiology challenge studies scope. Additionally, we observed wild-type pathogen.

Listeria monocytogenes is an intracellular, Gram-positive, non-spore-forming, non-encapsulated, facultative anaerobic, rod-shaped, and psychrotrophic food-borne pathogen that causes the infection, listeriosis, thus it attracts great attention following listeriosis outbreaks, which are often associated with high mortality rates. The prevalence of quite low globally; however, most recent deadliest outbreak occurred in South Africa, during 216 persons lost their lives. L. endowed potential to multiply through a wide range harsh environmental conditions, forming biofilms on varying surfaces food industry, as well having persistent antibiotic-resistant cells, pose major threat burden ready-to-eat industry. A more frustrating characteristic this bacterium its strain divergence, alongside increased level antibiotic resistance registered among strains recovered from food, humans, sources, especially those antibiotics involved treatment human listeriosis. Antibiotic exerted by pathogenic microbes ongoing public health menace continues be issue. Against background, thorough search into different databases using various engines was performed, led gathering salient information organised, chronologically, based Altogether, findings elaborated study present up-to date knowledge aspects will improve our understanding mystery ways prevent control dissemination foods. In addition, constant monitoring profiles sources detected changes, giving update trend resistance. Overall, bacterial contamination serves key aspect safety output
