This study aimed to evaluate occurrence of L. monocytogenes obtained from environment Czech Republic (hereinafter Czechia) and Slovakia 2016–2018 compare clonal relationship strains circulating with originating food chain humans. Altogether, 217 samples mud, surface water, vegetation soil were collected 61 locations. Samples processed according modified EN ISO 11290-1 standard. The isolates characterised using serotyping, macrorestriction analysis, followed by pulsed-field gel electrophoresis, whole genome sequencing (WGS), antimicrobial susceptibility testing. Listeria monocytogenes is a saprophytic gram-positive bacterium and opportunistic foodborne pathogen that can cause listeriosis in humans. The incidence of has been rising globally and, despite antimicrobial treatment, the mortality rates associated with most severe forms such as sepsis, meningitis meningoencephalitis remain high. notification humans mandatory Lithuania, up to 20 cases are reported annually. However, no studies have described detailed virulence susceptibility profiles any clinical L. strains Lithuania. Accordingly, this study aimed describe antibiotic invasive perform in-depth characterization isolated from patients neuroinfections through whole-genome sequencing. A total 70 isolates were collected, mostly infected aged 65 or older, between 2016 2021 : 41 (58.6%) blood, 19 (27.1%) cerebrospinal fluid, 5 (7.1%) wounds, 1 (1.4%) pleural fluid brain abscess. Two phylogenetic lineages identified—I ( n = 16/70, 22.9%) II 54/70, 77.1%)—along three serogroups—IIa 53/70, 75.7%), IVb 22.9%), IIc 1/70, 1.4%). Genomic analysis showed high level diversity seven genotypes: ST6 6), ST155 5), ST8 4), ST504 2) singletons for ST37, ST451 ST2. Phylogenetic clustered these into two clades defined by serogroups IIa. Notably, five tightly together (difference 6–48 core SNPs reference 0, 4 44 each other) ST155, previously European outbreak. Ruminant meat is considered to be a potent vehicle of foodborne pathogen transmission. Edible coatings are promising for enhancing safety. Here, edible chitosan membranes were applied whole cuts beef and mutton test the survival pathogenic bacteria Listeria monocytogenes methicillin-resistant Staphylococcus aureus. Meat pieces weighing approximately 200 g inoculated with cocktail three S. aureus isolates or four L. (6.00 log10CFU/g). The encased in coating formed by immersion incubated aerobically vacuum-packaged LDPE/PA/LDPE bags up 21 days. A decrease both (x = −1.95 log10CFU/g, standard error 0.23 log10CFU/g) counts −1.07 0.26 was observed. No significant differences observed between monocytogenes-spiked pieces; statistically higher versus under similar treatments. Aerobic storage enhanced antibacterial effects chitosan—a trend that not According results, effective controlling growth monocytogenes.

The objectives of this study were, firstly, to compare a conventional (i.e., chlorinated alkaline) versus an alternative (chlorinated alkaline plus enzymatic) treatment effectivity for the elimination biofilms from different L. monocytogenes strains (CECT 5672, CECT 935, S2-bac and EDG-e). Secondly, evaluate cross-contamination chicken broth non-treated treated formed on stainless steel surfaces. Results showed that all were able adhere develop at approximately same growth levels (≈5.82 log CFU/cm2). When put into contact with model food, obtained average transference rate potential global 20.4%. Biofilms detergent rates similar as high number residual cells around 4 5 Log CFU/cm2) present surface, except EDG-e strain which diminished 0.45%, was related protective matrix. Contrarily, shown not produce due its biofilm control (<0.50% transference) 935 had behavior. Therefore, changing more intense cleaning treatments in processing environments can reduce risk cross-contamination.

As a common foodborne pathogen, infection with L. monocytogenes poses significant threat to human life and health. The objective of this study was employ comparative genomics unveil the biodiversity evolutionary characteristics strains from different regions, screening for potential target genes mining novel genes, thus providing reference value specific molecular detection therapeutic targets strains. Pan-genomic analysis revealed that regions have open genomes, solid genetic basis adaptation environments. These contain numerous virulence contribute their high pathogenicity. They also exhibit relatively resistance phosphonic acid, glycopeptide, lincosamide, peptide antibiotics. results mobile elements indicate that, despite being located in geographical locations, there is certain degree similarity bacterial genome evolution environmental pressures. identified through pan-genomics are primarily associated fundamental activities invasion monocytogenes, including known such as inlB, which can be utilized purposes. After large number we further screened them using hub gene selection methods genes. present employed eight methods, ultimately identifying ten highly connected (bglF_1, davD, menE_1, tilS, dapX, iolC, gshAB, cysG, trpA, hisC), play crucial roles pathogenesis monocytogenes. pan-genomic showed evolution.
