Aims: Research on probiotics for constipation management is still growing, and plays a crucial role in the definition of strategy bowel wellbeing, constipation, related outcomes. The present systematic review meta-analysis beneficial effects SYNBIO® blend, to consolidate data from various clinical trials, was conducted. Methods: A literature search using PubMed, Web Science, Google Scholar databases limited trials that used either as dietary supplements or probiotic-enriched foods. Independently, two reviewers evaluated trial’s quality extracted all data. 95% confidence interval (CI) weighted mean difference (MD) pool continuous For analysis, Review Manager version 5.4 used. Results: Seven involving total 1095 subjects were included analysis. Overall, blend significantly improved relief by 0.75 (95% CI: 0.31 1.19; p = 0.0008) 52% subjects, increased intestinal regularity 1.90 compared placebo 1.02 2.78; < 0.0001) more than 60% individuals. No adverse events reported. Conclusions: found improve overall regularity, abdominal pain, cramping. This suggests people with these specific symptoms could benefit this probiotic combination.

Constipation is currently one of the most common gastrointestinal disorders, and its causes are diverse. Multi-strain probiotics often considered a more effective treatment than single-strain probiotics. In this study, constipation model was constructed using loperamide hydrochloride to evaluate ability multi-strain probiotic combination four different ratios Bifidobacterium Lactobacillus regulate intestinal flora, relieve constipation, explore initial mechanism in mice. After weeks intervention, BM1, BM2, PB2 effectively relieved constipation; however, pathways involved were different. The Bifidobacteria-dominated formulations BM1 BM2 mainly changed composition structure flora significantly decreased relative abundance Tyzzerella, Enterorhabdus, Faecalibaculum, Gordonibacter, Mucispirillum stool; increased Parabacteroides content short-chain fatty acids (SCFAs) restored motilin (MTL) vasoactive peptide (VIP) levels; downregulated interleukin 6 (IL-6) IL-8 levels serum. This repaired inflammatory response caused by constipation. Finally, it promoted peristalsis tract, increasing stool water content, relieving While Lactobacillus-dominated formula serum neurotransmitters (MTL, SP (substance P), VIP PYY (Peptide YY)) factors (IL-1, IL-6 IL-8), Gordonibacter then acetic acid thereby reducing level inflammation changing properties motility.

Abstract Diarrhea, a common gastrointestinal symptom in health problems, is highly associated with gut dysbiosis. The purpose of this study to demonstrate the effect multistrain probiotics (Sensi-Biome) on diarrhea from perspective microbiome-neuron axis. Sensi-Biome ( Lactiplantibacillus plantarum , Bifidobacterium animalis subsp. lactis Lactobacillus acidophilus Streptococcus thermophilus bifidum and Lactococcus ) was administered 4% acetic acid–induced rat model at concentrations 1 × 10 8 (G1), 9 (G2), CFU/0.5 mL (G3). Diarrhea-related parameters, inflammation-related cytokines, stool microbiota analysis by 16S rRNA were evaluated. A targeted untargeted metabolomics approach used analyze cecum samples using liquid chromatography orbitrap mass spectrometry. moisture content p < 0.001), intestinal movement rate 0.05), pH 0.05) significantly recovered G3. Serotonin levels decreased groups. inflammatory serotonin, tryptophan hydroxylase expression improved At phylum level, showed highest relative abundance Firmicutes . Short-chain fatty acids including butyrate, iso-butyrate, propionate, iso-valeric acid modified Equol oleamide In conclusion, effectively controls modulating metabolites serotonin pathway.

Antiretroviral therapy (ART) effectively reduces opportunistic infections and mortality in people living with HIV (PLWH); however, some patients exhibit poor immune recovery. This study explores the connections among responses, metabolites, gut microbiota PLWH differing reactions to ART. We analyzed composition, markers 38 who showed an immunological response (IR) 32 did not (INR), as classified according CD4+ T-cell levels after 24 months of Additionally, vitro assays using cell counting kit 8, flow cytometry, quantitative real-time reverse transcription PCR were employed assess effects metabolites on viability, marker expression, cytokine levels. Gut metabolic profiles differed significantly between IR INR groups. Enterococcus was more abundant group, whereas [Ruminococcus]_gnavus_group reduced. Significant pathway alterations included decreased folate biosynthesis biotin metabolism. observed negative associations Parabacteroides activation T-cells, positive correlations CD4/CD8 ratios. inverse relationships these markers. Indole-3-acetyl-beta-1-D-glucoside (area under curve value = 0.8931), had best discriminatory ability. Further experiments that proportions CD4+CD57+, effector CD4+, CD4+PD1+, CD8+CD57+, CD8+, CD8+HLA-DR+ T cells. Moreover, mRNA expression analysis treatment led a suppression pro-inflammatory cytokines. The multi-omics approach highlighted potential biomarkers for recovery HIV, suggesting avenues further research into strategies.

The pathophysiology functional constipation (FC) associated reduction in mucin-2 (MUC2) secretion microbial dysbiosis. To investigate underlying mechanism RCNSP against FC through MUC2 gut mucosal microbiota. Ultra-performance liquid chromatography tandem mass spectrometry characterized composition elucidate material basis action. model was induced via loperamide gavage (16 mg/kg) twice daily 7 days. Applying defecation function gastrointestinal motility assess severity. Hematoxylin eosin Alcian blue-periodic acid-schiff staining analyzed colonic morphology. Opioid receptor agonists, particularly those that activate µ-opioid receptors (MORs), are essential analgesic agents for acute or chronic mild to severe pain treatment. However, their use has raised concerns including, among others, intestinal dysbiosis. In addition, growing data on constipation-evoked dysbiosis have been reported. Opioid-induced constipation (OIC) creates an obstacle continuing treatment with opioid analgesics. When non-opioid therapies fail overcome the OIC, antagonists peripheral, fast first-pass metabolism, and gastrointestinal localized effects remain drug of choice which discussed here. At first glance, seems only be restricted constipation, however, recent OIC-related its contribution appearance several side garnered a great attention from researchers. Peripheral MORs also considered as future target analgesics limited central effects. The properties MOR counteracting influence possibly peripheral MOR-mediated antinociception, will highlighted. A new concept is proposed developing gut-selective treat restore OIC while keeping antinociception unaffected. impact in relation changes gut microbiome included.
