Cryo multi-scale microscopy visualization tools for studying microbial communities on glacier surfaces

In
recent
decades,
a
growing
body
of
research
has
focused
on
characterizing
supraglacial
microbial
communities.
These
studies
have
unveiled
that
pigmented
glacier
ice
algae,
namely,
Ancylonema
nordenskiöldii
and
alaskanum
,
blooming
during
the
summer
melt
seasons
accelerate
surface
melting.
Most
techniques
quantify
processes
glacial
surfaces
require
melting
samples
prior
to
their
analysis
and,
so
far,
structure
three-dimensional
arrangement
microorganisms
bloom
bare
never
been
characterized.
To
address
this
gap,
we
developed,
tested
validated
first
characterization
workflow
for
imaging
colonizing
frozen
Greenland
Ice
Sheet.
While
preserving
state,
employed
suite
microscopic
tools
with
progressively
increasing
resolution,
starting
from
algae
hand-held,
portable
microscopes
(up
×150
magnification)
all
way
high-resolution
cryo-scanning
electron
microscopy
(cryo-SEM,
up
×5.000
magnification).
Our
images
provided
us
visualization
distribution,
structure,
interactions,
relationship
between
microbes,
minerals,
substrate.
innovative
approach
significantly
advances
understanding
life
within
matrix,
shedding
light
spatial
architecture
cells
in
on-ice
thereby
improving
how
perceive
study
these
unique
ecosystems.

stable
at
temperatures
up
70
°C,
achieving
activity
level
16.23
±
0.22
mm.
Furthermore,
maintains
its
efficacy
across
pH
range
2
12.
Notably,
diameter
recorded
16.53
0.12
mm
following
80
min
UV
irradiation.
Under
different
treatment
conditions,
fermentation
