One pair of novel enantiomers, gigantdioxin A (+)-1 and B (−)-1, with a skeleton benzo[d][1,3]dioxin; new acetophenone gigantone (3); known 3-chlorogentisyl alcohol (2), which is the bioprecursor 1; (4); chromone derivative (5) were obtained from sponge-associated fungus Aspergillus giganteus MA46-5. Their structures established by extensive in-depth spectral analysis, such as UV, 1D 2D NMR, HRESIMS. The absolute configurations (±)-1 deduced ORD, chiral separation, experimental computational ECD. Meanwhile, we proposed possible biosynthetic pathway (±)-1. Fortunately, was proved biomimetic synthesis through 2, bioprecursor, reacted n-butyraldehyde. Myocardial protection assays showed that 3 4 possessed stronger protective effects than positive control against myocardial cell H9c2 ischemia–reperfusion injury in low concentrations, effect (−)-1 almost equal to control. Further, explored mechanism network pharmacology. Adenosine A2a receptor (ADORA2A) serum albumin (ALB) represent potential targets for associated 4, respectively. Based on pharmacology, docked predicted proteins bioactive compounds using Autodock Vina.

Certain mangrove endophytic fungi have unique physiological mechanisms, such as anti-inflammatory properties. A new polyketone, namely N-acetoxyethyl-3-(3,5-dimethyl-1,3-heptadienyl)-5-chloro-7-methyl-7-acetoxy-6,8-(2H,7H)-isoquinolinedione (1), and 17 known compounds (2-18) were isolated from the fungus Diaporthe pseudooculi HHUF 30617. The structure of compound was elucidated using nuclear magnetic resonance (NMR) spectroscopy high-resolution electrospray ionisation mass (HRESI-MS). activities evaluated zebrafish migratory fluorescent cell count assay. results suggest potential above for treating inflammation. These findings aid development natural marine products based on fungal metabolites to counter conditions copper toxicity.

Thermophilic microorganisms possess several adaptations to thrive in high temperature, which is reflected as biosynthesis of proteins and thermostable molecules, isolation culture represent a great methodological challenge, therefore High throughput sequencing enables screening the whole bacterial genome for functional potential, providing rapid cost-effective information guide targeted cultures identification characterization novel natural products. In this study, we isolated two thermophilic strains corresponding Bacillus LB7 Streptomyces LB8, from microbial mats Atacama Desert. By combining mining, biochemical characterization, aimed identify their capacity synthesize bioactive compounds with antimicrobial properties. Additionally, determined capability produce under controlled vitro assays detected by determining masses Thin-Layer Chromatography/Mass Spectrometry (TLC/MS). Overall, both isolates can (e.g., Myxalamide C by-product) antioxidants (e.g. Dihydroxymandelic Acid, Amide biotine Flavone by-products) compounds. strain possesses more diverse repertoire 51.95% total metabolites unmatched, while LB8 favors mainly antioxidants, but has over 70% unclassified compounds, highlighting necessity study elucidate structure Based on these results, postulate that uncultured or rare cultured thermophiles inhabiting high-altitude hydrothermal ecosystems Desert offer promising opportunity
