Air-curing is the initial step in processing of cigar tobacco leaves. However, dynamics microbial community and metabolic functions different parts leaves during this process remain largely unclear. In study, amplicon-based high-throughput sequencing revealed that Pseudomonas (9.0 to 29.9%) Sphingomonas (0.5 13.8%) were dominant bacterial genera early stages air-curing, while Pantoea (1.7–90.4%) became predominant after air-curing. Microbial diversity analysis indicated species richness significantly higher fresh leaf withering periods. Functional prediction based on PICRUSt2 suggested communities middle exhibited abundances pathways related carbohydrates amino acids than those upper leaves, potentially leading formation more flavor compounds. The volatile compounds detected air-curing by HS-SPME-GC–MS, with alkaloids esters being most prominent, although their accumulation periods differed across parts. To study the composition and succession of bacterial fungal communities during fermentation cigar filler leaves with varying initial water contents, high-throughput sequencing technology was used to sequence 16SrRNA genes ITS1 from tobacco leaf samples. This followed by analyses microbial α-diversity, community structure, function prediction based on data. The diversity richness decreased over time under different content conditions. Among 18 samples, predominant phyla identified were Proteobacteria, Firmicutes, Actinobacteria, Ascomycota, Basidiomycota, leading genera being Staphylococcus, Sphingomonas, Methylobacterium-Methylorubrum, Pseudomonas, Humicola. Functional predictions for bacteria revealed their primary involvement in carbohydrate, lipid, amino acid metabolism. influenced structure relative abundance fermentation. While exhibited a similar structural composition, there notable differences abundance. functional results PICRUSt indicated that predicted species among samples minimal, whereas variations more pronounced across stages contents.
