Air-curing is the initial step in processing of cigar tobacco leaves. However, dynamics microbial community and metabolic functions different parts leaves during this process remain largely unclear. In study, amplicon-based high-throughput sequencing revealed that Pseudomonas (9.0 to 29.9%) Sphingomonas (0.5 13.8%) were dominant bacterial genera early stages air-curing, while Pantoea (1.7–90.4%) became predominant after air-curing. Microbial diversity analysis indicated species richness significantly higher fresh leaf withering periods. Functional prediction based on PICRUSt2 suggested communities middle exhibited abundances pathways related carbohydrates amino acids than those upper leaves, potentially leading formation more flavor compounds. The volatile compounds detected air-curing by HS-SPME-GC–MS, with alkaloids esters being most prominent, although their accumulation periods differed across parts. Furthermore, PLS-DA, 17 38 changed components identified respectively. Finally, potential relationships between characteristic microbes explored Spearman correlation coefficient. It was found multiple bacteria such as Rhodanobacter , Gemmatimonas Ramlibacter present significant positive correlations 3,3-dimethylacrylic acid, phenylacetone, 2,3-butanedione, geranylacetone, promoting process. This study provides scientific insights into role microorganisms offers a basis for screening specific functional improve stabilize future.
