Phosphorylated metabolites, here referred to as phosphometabolites, are sufficiently abundant and widely distributed provide a condensed representation of metabolism that can be readily accessed through NMR spectroscopy. This study addresses the challenge precisely quantifying phosphometabolites via quantitative 31P from tissue extracts. We optimized standard operating procedures for enhanced spectral resolution, signal intensity, accuracy. By amply evaluating solvent buffer conditions, reference compounds, paramagnetic relaxation enhancers, we identified optimal conditions metabolite analysis, including use trimethylphosphine oxide accurate referencing due its short T1 time minimal offset glycine (in D2O) at pD 9.5, where virtually invariant frequencies sample osmolarities observed, along with maximal detection sensitivity temperature stability pH. To study the differences in urine metabolome between pediatric patients with severe Mycoplasma pneumoniae pneumonia (SMPP) and those general (GMPP) via non-targeted metabolomics method, potential biomarkers were explored through machine learning (ML) algorithms. The metabonomics data of 48 children SMPP 85 GMPP collected high performance liquid chromatography‒mass spectrometry (HPLC-MS/MS). differential metabolites two groups obtained principal component analysis (PCA) partial least squares-discriminant (PLS-DA), significant metabolic pathways screened enrichment analysis.
