Lead (Pb) is a common pollutant that not biodegradable and gravely endangers the environment human health. Annona squamosa fruit has wide range of medicinal uses owing to its phytochemical constituents. This study evaluated effect treatment with A. extract (ASFE) on testicular toxicity induced in male rats by lead acetate. The metal-chelating capacity composition ASFE were determined. LD50 was probit analysis. Molecular docking simulations performed using Auto Dock Vina. Forty Sprague Dawley equally divided into following groups: Gp1, negative control group; Gp2, given (350 mg/kg body weight (b. wt.)) (1/10 LD50); Gp3, acetate (PbAc) solution (100 b. wt.); Gp4, PbAc as Gp3 Gp2. All treatments oro-gastric intubation daily for 30 days. Body changes, spermatological parameters, reproductive hormone levels, oxidative stress inflammatory biomarkers evaluated, molecular histopathological investigations performed. results showed had promising activity composition. 3500 wt. analysis quercetin demonstrated high binding affinity JAK-1 STAT-3 proteins, this could make it more candidate targeting JAK-1/STAT-3 pathway than others. defective tissues, altered molecular, biochemical, histological features, well impaired characteristics. Treatment led significant mitigation these dysfunctions modulated JAK-1/STAT-3/SOCS-1 axis rats.

Abstract Doxorubicin (DOX) is used to treat a variety of malignancies; however, its cardiotoxicity limits effectiveness. Shikimic acid (SA) showed several promising biomedical applications. This study investigated the protective effect SA on DOX-induced in male rats. The ADMETlab 2.0 web server was predict pharmacokinetic properties SA. Molecular docking studies were conducted using AutoDock Vina. Fifty rats divided into 4 groups ( n = 10); G1 negative control; G2 injected with mg/kg DOX intraperitoneally (i.p.) once week for month; G3 gavaged by 1/10 LD 50 (280 mg/kg) daily month, and G4 as G3. After hematological, biochemical, molecular, histopathological investigations assessed. results that treatment led significant amelioration restoring inflammatory biomarkers, antioxidant gene expression, cardiac alterations. Importantly, impact against DOX-promoted deterioration targeting Nrf-2/Keap-1/HO-1/NQO-1 signaling pathway, which turn induces agents. These findings suggest could potentially mitigate toxicity during DOX-based chemotherapy.

Osteocyte senescence is associated with skeletal dysfunction, but how to prevent bone loss and find the effective therapeutic targets a potential scientific concern. Cadmium (Cd) widespread environmental contaminant that causes substantial damage in both animals humans. Oligomeric proanthocyanidins (OPC) are naturally polyphenolic substances found various plants demonstrate significant anti-senescence potential. Here, we investigated protective effects of OPC against Cd-induced osteocytes identify regulatory mechanisms. alleviated by attenuating cell cycle arrest, reducing ROS accumulation, suppressing pro-inflammatory responses vitro. Furthermore, effectively prevented breakdown dendritic synapses Correspondingly, ameliorated through activity vivo. Taken together, our results establish as promising agent ameliorates osteocyte mitigating oxidative stress inflammatory responses.
