Fucosidosis is a rare lysosomal storage disease caused by α-L-fucosidase deficiency following mutation in the FUCA1 gene. This enzyme responsible for breaking down fucose-containing glycoproteins, glycolipids, and oligosaccharides within lysosome. Mutations result either reduced activity or complete loss of function, leading to accumulation fucose-rich substrates lysosomes. Lysosomes become engorged with undigested substrates, which leads secondary defects affecting other metabolic pathways. The central nervous system particularly vulnerable, dysfunction causing microglial activation, inflammation, neuronal loss, neurodegenerative symptoms fucosidosis. Neuroinflammation contributes damage, including apoptosis, axonal degeneration, synaptic dysfunction, exacerbating process. Chronic neuroinflammation impairs plasticity survival, progressive intellectual disability, learning difficulties, previously acquired skills. Inflammatory cytokines burden motor neurons associated pathways contribute ataxia, spasticity, hypotonia, are common Elevated neuroinflammatory markers can increase excitability, frequent occurrence epilepsy affected individuals. So, fucosidosis characterized rapid mental along growth retardation, coarse facial features, hepatosplenomegaly, telangiectasis angiokeratomas, epilepsy, inguinal hernia, dysostosis multiplex. Patients usually die at an early age. Treatment great challenge, there currently no definitive effective treatment. Hematopoietic cell transplantation studies ongoing treatment However, diagnosis this be effective. In addition, body’s immune decreases due chemotherapy applied after transplantation, leaving body vulnerable microbes infections, risk death high another method, gene therapy, use retroviral vectors, promising their easy integration, efficiency, safety. approach, replacement preclinical fucosidosis, but blood–brain barrier major obstacle diseases system. Abstract Background N-glycosylation is a complex, post-translational modification which influences protein function and sensitive to physiological changes. Obesity associated with alterations in function; however, little known about the glycoproteome obesity beyond observations of association types structures selected glycopeptides. Most often, due technical challenges, glycan composition structure information are missing. Here, we combined label-free data-independent proteomics targeted quantitative glycoproteomics study plasma proteins obesity. Using monozygotic twin design, controlled for genetic variation focused only on acquired effects Methods samples 48 pairs discordant BMI (intrapair difference > 2.5 kg/m 2 ), identified using mass spectrometry, differential glycopeptide levels between heavier leaner co-twins. We used within-twin paired analysis model considered p < 0.05 as significant. Results 33 expression differences ( 0.05) These occurred either both glycoprotein (sometimes opposing directions) or independently from each other. Haptoglobin was upregulated (Fold Change = 1.10, 0.001) co-twins along seven compositions at site Asn241. The complement C3 1.08, 0.014) one Asn85. Additionally, many glycopeptides were despite non-significant protein-backbone levels. The brain contains many interconnected and complex cellular molecular mechanisms. Injury to the causes permanent dysfunctions in these So, it continues be an area where surgical intervention cannot performed except for removal of tumors repair some aneurysms. Some agents that can cross blood–brain barrier reach neurons show neuroprotective effects due their anti-apoptotic, anti-inflammatory antioxidant properties. In particular, act by reducing or modulating accumulation protein aggregates neurodegenerative diseases (Alzheimer’s disease, Parkinson’s Huntington’s Amyotrophic lateral sclerosis, prion disease) caused accumulation. Substrate increased oxidative stress stimulates brain’s immune cells, microglia, astrocytes, secrete proinflammatory cytokines. Long-term chronic neuroinflammatory response triggers apoptosis. Brain damage is observed with neuronal apoptosis functions are impaired. This situation negatively affects processes such as motor movements, memory, perception, learning. Neuroprotective prevent molecules play a role addition, they improve impaired supporting neuroplasticity neurogenesis. Due important roles central nervous system diseases, elucidate review provides overview mechanisms flavonoids, which constitute large part effects, well vitamins, neurotransmitters, hormones, amino acids, derivatives. Human milk oligosaccharides (HMOs), the third most abundant solid component in human milk, vary significantly among women due to factors such as secretor status, race, geography, season, maternal nutrition and weight, gestational age, delivery method. In recent studies, HMOs have been shown a variety of functional roles development infants. Because are not digested by infants, they act metabolic substrates for certain bacteria, helping establish infant’s gut microbiota. By encouraging growth advantageous intestinal these sugars function prebiotics produce short-chain fatty acids (SCFAs), which essential health. can also specifically reduce harmful microbes viruses binding epithelium, preventing illness. HMO addition infant formula is safe promotes healthy development, infection prevention, Current formulas frequently contain (OSs) that differ structurally from those found making it unlikely would reproduce unique effects HMOs. However, there growing trend producing OSs resembling HMOs, but limited data make unclear whether offer additional therapeutic benefits compared non-human OSs. Better knowledge how mammary gland synthesizes could direct technologies yield broad complex with OS compositions closely mimic milk. This review explores HMOs’ nature vital role health, examining variation composition its contributing factors. It highlights technological advances enabling large-scale studies on Furthermore, multifunctional biological processes brain microbiota immune response regulation investigated. Breast milk oligosaccharides (BMOs) are short polymers of simple sugars that present in high concentrations a woman’s breast (BM). Their content changes fast over time. Genetic factors, the duration lactation, and environmental factors particularly influence level BMOs. BMOs can facilitate development immune system protect against infectious diseases directly through interaction intestinal epithelial cells or indirectly modulation microbiota, including stimulating Bifidobacterium growth. Clinical data suggest adding to infant formula promotes normal neonatal growth benefits health. Adding 1 2 formulas is safe brings their composition closer BM, allowing child develop harmoniously.

Bovine colostrum (BC), the first milk secreted by mammals after birth, is a trending alternative source for supplementing infants and children, offering benefits gut immune health. Its rich components, such as proteins, immunoglobulins, lactoferrin, glycans, are used to fortify diets support development. Preterm development crucial, especially in maturation of essential systems, from 2010 2020, approximately 15% all premature births occurred at less than 32 weeks gestation worldwide. This review explores composition, benefits, effects BC on general along with preterm who require special care, highlights its role growth also associated specific pediatric diseases, including necrotizing enterocolitis (NEC), infectious diarrhea, inflammatory bowel disease (IBD), short-bowel syndrome (SBS), neonatal sepsis, gastrointestinal respiratory infections, some minor conditions. discusses clinical trials regarding these conditions which occasionally encountered infants. The anti-inflammatory, antimicrobial, immunomodulatory, antiviral properties discussed, emphasizing mechanisms action. Clinical trials, particularly humans, provide evidence supporting inclusion formulas diets, although precise standards age, feeding time, amounts needed ensure safety efficacy. However, potential adverse effects, allergic reactions caseins immunoglobulin E, must be considered. More comprehensive necessary expand infant feeding, important components BC, should further studied their synergistic diseases.
