In Parkinson's disease, dopaminergic neurons located in the substantia nigra of brain are destroyed, affecting patients' motor function. The endocannabinoid system is responsible for controlling neuronal homeostasis and its alteration related to neurodegenerative diseases, such as Parkinson's. A literature review regarding relationship between disease was carried out through a search Pubmed database. Complete publications from last year were included, using 15 papers. Treatment with cannabinoid medications patients should not be first choice, being restricted adjuvant therapy they elderly vulnerable. Cannabis extracts have shown experimental studies neuroprotective inflammation modulating actions. Pure cannabidiol safe, few side effects. Computational analyzes demonstrated that binding CB1 CB2 receptors induced structural changes them. Levels lower specific areas decline these correlated worse severity symptoms. Selective receptor agonists immunomodulatory actions, reducing formation defective proteins. Hyperpolarized current-induced inhibition improved muscle stiffness but worsened symptoms depression anxiety animals. use exogenous cannabinoids yet consolidated, more clinical needed confirm safety interaction system.

The endocannabinoid system (ECS), named after the chemical compounds found in cannabis plant, is a regulatory network of neurotransmitters, receptors, and enzymes that plays crucial roles skin health disease. Endogenous ligands ECS, called endocannabinoids, have proven to be important regulators immune responses. One most prevalent arachidonoylethanolamide (also known as anandamide), for its anti-inflammatory effects. Langerhans cells (LCs) are sole antigen-presenting present human epidermis. They serve first line defense against pathogens essential skin's specific responses play critical role maintaining tissue homeostasis; however, little about effect endocannabinoids on these cells. Our research aimed provide connection between monocyte-derived (moLCs) shedding light their collaborative homeostasis inflammation. Human monocytes were differentiated into moLCs using established protocols. Anandamide was applied during differentiation process test viability, marker expression, cytokine production cells, well short term treatments intracellular calcium measurement. TLR protocol used activate moLCs. impact anandamide functionality further assessed differential gene expression analysis bulk RNA-Seq data, moLC-T cell cocultures, while ELISpot employed determine polarization T activated aforementioned cocultures. did not significantly affect viability up 10 µM. When it had only negligible CD207 prototypic LCs; there an observed reduction CD1a by no significant effects maturation status moLCs, nor induced TLR3 TLR7/8 agonists. MoLCs presence however show decreased CXCL8, IL-6, IL-10 IL-12 cytokines activation. Anandamide-treated showed increased capability naïve cells; level seen with combined agonism. RNA sequencing modest changes compared control but reveal inhibitory oxidative phosphorylation specifically also promoted towards Th1 phenotype. results has nuanced differentiation, maturation, secretion, metabolism function Among decrease holds promise selectively dampen inflammation restricted which been implicated drivers common inflammatory conditions such psoriasis, atopic dermatitis contact dermatitis.

Skin is the largest organ of human body and undergoes both intrinsic (chronological) extrinsic aging. While skin aging driven by genetic epigenetic factors, mediated external threats such as UV irradiation or fine particular matters, sum which referred to exposome. The clinical manifestations biochemical changes are different between aging, albeit overlapping features exist, eg, increased generation reactive oxygen species, extracellular matrix degradation, telomere shortening, lipid peroxidation, DNA damage. As a prominent target for many hormones, molecular processes underlying under tight control classical neuroendocrine axes. However, also an endocrine itself, including hair follicle, fully functional "miniorgan." Here we review pivotal hormones controlling focusing on IGF-1, key fibroblast-derived orchestrator GH, estrogens, retinoids, melatonin. emerging roles additional players, ie, α-melanocyte-stimulating hormone, central player hypothalamic-pituitary-adrenal axis; members hypothalamic-pituitary-thyroid oxytocin, endocannabinoids, peroxisome proliferator-activated receptor modulators, reviewed. Until now, only limited number these mainly topical retinoids have found their way into practice anti-skin compounds. Further research biological properties players its derivatives may offer development novel senotherapeutics treatment prevention
