Background: Pain is a noxious, sensory, and emotional experience due to intense stimuli. intimately linked with nociception. The clinical therapies available at the moment for pain are tricyclic antidepressants, non -steroidal anti-inflammatory drugs, opioids. There need develop investigate safer alternatives that minimise chances of psychological or physical drug dependence dose tolerance. purpose this research an anti-nociceptive formulation (Markhamia tomentosa cream co-formulated Eucalyptus oil as permeation enhancer) evaluate its efficacy. Methods: In study, Markhamia was extracted from leaves using maceration technique. A qualitative screening ethanolic extract (EEMT) carried out. Five emulsions (A1-A5) containing varying compositions were developed, negative control (an emulsion did not contain oil) also developed fusion method. properties topical creams evaluated mice paw-licking xylene-induced ear oedema tests. Diclofenac aspirin administered 20 mg/kg 50mg/kg orally, respectively, used standards. All experiments out in triplicates. Metadata is essential for handling medical data according to FAIR principles. Standards are well-established many types of electrophysiological methods but still lacking microneurographic recordings peripheral sensory nerve fibers in humans. Developing a new concept enhance laboratory workflows complex process. We propose standard structuring and storing microneurography metadata based on odML odML-tables. Further, we present an extension the odML-tables GUI that enables user-friendly search functionality database. With our open-source repository, encourage other labs incorporate odML-based into their experimental routines.

Abstract Spike sorting represents a persistent challenge in electrophysiology, particularly extracellular nerve recordings containing signals from several fibers. This issue is exacerbated microneurography peripheral unmyelinated afferents awake humans, which are responsible for pain sensation. due to the similarity of spike shapes originating different fibers, low signal-to-noise ratios, and shape-distorting overlaying signals. Here, we present first systematic assessment morphology-based multiple two laboratories. We created dedicated ground truth datasets by employing semi-manual labelling methods enabling comparison supervised unsupervised feature sets. A strong advantage approach was observed, while no single set showed global advantage. Further, high diversity results linked per-recording fiber number morphologies. To extend this problem microneurography, our open-source pipeline enables reproducible sortability analysis any neuronal activity if electrical stimulation fibers possible. The achieved advancement lays foundation gaining insights into neural coding itch clinical context.
