The identification of safe and easily-determined-inflight biomarkers to monitor the immune system astronauts is mandatory ensure their well-being success missions. In this report, we evaluated relevance two whose determination could be easily implemented in a spacecraft near future by using bedridden volunteers as ground-based model microgravity spaceflight. Our data confirm neutrophil lymphocyte ratio (NLR) suggest platelet (PLR) monitoring assess long-lasting diseases. We recommend coupling these ratios other biomarkers, such quantification cytokines viral load measurements, efficiently detect dysfunction, determine when countermeasures should applied promote recovery, prevent development disease, track responses treatment.

Aging induces numerous physiological alterations, with immunosenescence emerging as a pivotal factor. This phenomenon has attracted both researchers and clinicians, prompting profound questions about its implications for health disease. Among the contributing factors, one intriguing actor in this complex interplay is human cytomegalovirus (CMV), member of herpesvirus family. Latent CMV infection exerts influence on aging immune system, potentially to age-related diseases. review delves into intricate relationship between CMV, revealing how chronic viral impacts landscape. We explore mechanisms through which can impact composition functionality cell populations induce shifts inflammatory profiles aging. Moreover, we examine potential role pathologies such cardiovascular diseases, cancer, neurodegenerative disorders, COVID-19, Long COVID. underlines importance understanding CMV. Multiple sclerosis (MS) primarily affects adult females. However, in the last decades, rising incidence and prevalence have been observed for demographic extremes, such as pediatric-onset MS (POMS; occurring before 18 years of age) late-onset (corresponding to an onset above 50 years). These categories show peculiar clinical-pathogenetic characteristics, aging processes disease courses, therapeutic options, unmet needs. Nonetheless, several open questions are still pending. POMS patients display important contribution multiple genetic environmental factors EBV, while LOMS, hormonal changes pollution may represent triggers. In both categories, immunosenescence emerges a pathogenic driver disease, particularly LOMS. populations, patient caregiver engagement essential from diagnosis communication early treatment disease-modifying therapy (DMTs), which elderly population appears more complex less proven terms efficacy safety. Digital technologies (e.g., exergames e-training) recently emerged with promising results, treating following motor cognitive deficits. this offer seems feasible POMS, being LOMS familiar digital technology. narrative review, we discuss how process influences pathogenesis, course, options Finally, evaluate impact new tools, greatly interest current future management patients.

The process of aging is accompanied by a dynamic restructuring the immune response, phenomenon known as immunosenescence. This mini-review navigates through complex landscape age-associated changes, chronic inflammation, age-related autoimmune tendencies, and their potential links with immunopathology Long COVID. Immunosenescence serves an introductory departure point, elucidating alterations in cell profiles functional dynamics, changes T-cell receptor signaling, cytokine network dysregulation, compromised regulatory function. Subsequent scrutiny or “inflammaging,” highlights its roles susceptibilities mediator perturbations observed COVID patients. introduction epigenetic facets further amplifies interconnections. In this compact review, we consider interactions between immunosenescence, autoimmunity. We aim to explore multifaceted relationships that link these processes shed light on underlying mechanisms drive interconnectedness. With focus understanding immunological context aging, seek provide insights into how immunosenescence inflammation contribute emergence progression disorders elderly may serve for disturbances.

Abstract Renal aging, marked by the accumulation of senescent cells and chronic low‐grade inflammation, leads to renal interstitial fibrosis impaired function. In this study, we investigate role macrophages, a key regulator in aging analyzing kidney single‐cell RNA sequencing data C57BL/6J mice from 8 weeks 24 months. Our findings elucidate dynamic changes proportion cell types during reveal that increased macrophage infiltration contributes with these macrophages exhibiting senescence activation ferroptosis signaling. CellChat analysis indicates enhanced communications between tubular aging. Suppressing alleviates macrophage‐mediated partial epithelial‐mesenchymal transition vitro, thereby mitigating expression fibrosis‐related genes. Using SCENIC analysis, infer Stat1 as age‐related transcription factor promoting iron dyshomeostasis regulating Pcbp1, an chaperone protein inhibits ferroptosis. Furthermore, through virtual screening molecular docking library anti‐aging compounds, construct model targeting which natural small molecule compound Rutin can suppress preserving Pcbp1. summary, our study underscores crucial results also suggest Pcbp1 intervention target aging‐related highlight potential therapeutic agent inflammation fibrosis.

Aging is associated with a progressive decline of innate and adaptive immune responses, called immunosenescence. This phenomenon links to different multiple sclerosis (MS) disease courses among age groups. While clinical relapse active demyelination are mainly related the altered immunity, including invasion T- B-lymphocytes, impairment cell (e.g., microglia, astrocyte) function main contributor disability progression neurodegeneration. Most patients MS manifest relapsing-remitting phenotype at younger age, while phenotypes seen in older patients. Current disease-modifying therapies (DMTs) primarily targeting immunity less efficacious patients, suggesting that immunosenescence plays role treatment response. review summarizes recent mechanistic studies regarding discusses implications these findings.
