In Silico Insights into Micro-Mechanism Understanding of Extracts of Taxus Chinensis Fruits Against Alzheimer’s Disease DOI

Background: The taxus chinensis fruit (TCF) shows promises in treatment of aging-related diseases such as Alzheimer’s disease (AD). However, its related constituents and targets against AD have not been deciphered. Objective: This study was to uncover TCF extracts AD. Methods: An integrated approach including ultrasound extractions constituent identification by UPLC-QE-MS/MS, target R data-mining from Pubchem, Drugbank GEO databases, network construction, molecular docking the ROC curve analysis carried out. Results: We identified 250 compounds extracts, obtained 3,231 known 5,326 differential expression genes AD, 988 intersection genes. Through construction KEGG pathway analysis, 19 chemicals, 31 targets, 11 biological pathways were core compounds, Among these constituents, luteolin, oleic acid, gallic baicalein, naringenin, lovastatin rutin had obvious anti-AD effect. Molecular results further confirmed above results. AUC values about 87% greater than 0.5 two chips especially 10 with 0.7, BCL2, CASP7, NFKBIA, HMOX1, CDK2, LDLR, RELA, CCL2, which mainly referred neuron apoptosis, response oxidative stress inflammation, fibroblast proliferation, etc. Conclusions: diverse active that can act on diagnostic deserve in-depth study.

Abstract Improving the quality of life in elderly patients and finding new treatment options for neurological diseases such as Alzheimer’s has become one priorities scientific world. In recent years, beneficial effects therapeutic properties natural foods on health have a very remarkable issue. Walnut oil (WO) is promising nutraceutical, with many phytochemicals polyunsaturated fatty acids thought to be cognitive deficits, disease (AD). Polyphenolic compounds found WO enhance intraneuronal signaling neurogenesis improve sequestration insoluble toxic protein aggregates. The objective this study was investigate potential protective model AD induced by retinoic acid (RA) brain-derived neurotrophic factor (BDNF). order achieve this, experimental groups were formed follows: Control group, (AD) + applied group (AD WO). supplementation almost significantly reduced oxidative stress ad model, providing 2-fold protection against oxidation. Additionally, showed significant reduction tau levels (2-fold), increased acetylcholine (ACh) (12%), decreased esterase (AChE) activity (~50%). Since it been known centuries that does show any adverse human neuroprotective properties, may used an additional nutraceutical drug treatments.
