Fibrinogen, a pivotal plasma glycoprotein, plays an essential role in hemostasis by serving as the precursor to fibrin, which forms structural framework of blood clots. Beyond coagulation, fibrinogen influences immune responses, inflammation, and tissue repair. Oxidative stress, characterized imbalance between reactive oxygen species (ROS) antioxidants, induces oxidation, significantly altering its structure function. This narrative review synthesizes findings from vitro, ex vivo, clinical studies, emphasizing impact oxidation on clot formation, architecture, degradation. modifications result denser fibrin clots with thinner fibers, reduced permeability, heightened resistance fibrinolysis. These changes exacerbate prothrombotic conditions cardiovascular diseases, diabetes, chronic inflammatory disorders cancer. In contrast, “low-dose” oxidative stress may elicit protective adaptations fibrinogen, preserving The also highlights discrepancies experimental due variability protocols patient conditions. Understanding interplay function could unveil therapeutic strategies targeting stress. Antioxidant therapies or selective inhibitors detrimental hold potential for mitigating thrombotic risks. However, further research is pinpoint specific sites, clarify their roles dynamics, bridge gap basic practice.

Histidine is a versatile amino acid with metal-binding, nucleophilic, and basic properties that endow many peptides proteins biological activity. However, histidine itself susceptible to oxidative modifications via post-translational modifications, photo-oxidation, metal-catalyzed oxidation. Despite multiple investigations into these different oxidation systems, the varied attributions differential outcomes point significant gaps in our understanding of coordination requirements, spectral features, reaction products accompany Cu-catalyzed histidine-containing peptides. Here, we use model Histatin-5, salivary peptide Cu-potentiated antifungal activity relies on its residues, characterize complex mixture resulting from Cu under physiologically relevant reducing oxidizing conditions. Characterization LC-MS, MS/MS, UV-vis, NMR revealed adjacent residues bis-His site are main target oxidation, predominant being 2-oxo-His His-His cross-links give rise distinctive electronic absorption features between 300-400 nm. Doubly- triply-oxygenated peptides, intramolecular cross-links, multimers case shorter were also observed. The configuration motif may enable reactivity not available systems where His sequence or space. These results expand possibilities other containing histidines.

Alzheimer's disease (AD) and frontotemporal dementia (FTD) are the most common forms of globally. AD is characterized by accumulation amyloid-β (Aβ) plaques hyperphosphorylated tau in brain, leading to progressive memory loss cognitive decline, significantly impairing daily life. In contrast, FTD marked selective degeneration frontal and/or temporal lobes, typically resulting profound changes personality social behavior, speech disorders, psychiatric symptoms. Numerous studies have found microRNAs (miRNAs)-small, non-coding RNA molecules that regulate gene expression post-transcriptionally-to be dysregulated FTD. As a result, miRNAs emerged as promising novel biomarkers for these diseases. This review examines current understanding FTD, emphasizing their potential accessible, noninvasive diagnosing prevalent neurodegenerative disorders.

Abstract Within most tissues, the extracellular microenvironment provides mechanical cues that guide cell fate and function. Changes in matrix such as aberrant deposition, densification increased crosslinking are hallmarks of late-stage fibrotic diseases often lead to organ dysfunction. Biomaterials have been widely used mimic properties study However, initiation fibrosis has largely overlooked, due challenges recapitulating early lesions within native microenvironment. Using visible light mediated photochemistry, we induced local stiffening proteins ex vivo murine human tissue. In lung tissue epithelial lineage-traced mice, mimicked alveolar spreading, differentiation remodeling. inhibition cytoskeletal tension or integrin engagement reduced spreading differentiation, resulting dedifferentiation deposition. Our findings emphasize role remodeling early-stage implications for disease modeling applications other tissues.

Introduction: Bipolar affective disorder (BD), also known as manic-depressive illness, is a chronic and recurrent psychiatric characterised by significant mood disturbances. It one of the leading causes disability worldwide associated with high risk suicide. Recent studies highlight role oxidative stress (OS) in pathogenesis BD. The body's pro/antioxidant imbalance adversely affects cellular molecular processes. Aim: aim this review to synthesise current state knowledge on OS aetiology course BD, including key biomarkers potential therapeutic interventions. Methods: A scientific literature was conducted, articles published between 2000 2024. Searches were conducted PubMed, Scopus Web Science databases, using following keywords: 'bipolar disorder', 'oxidative stress', 'antioxidants', 'biomarkers', 'mitochondrial dysfunction', 'redox homeostasis', 'treatment'. Results: Results indicate that patients BD have elevated levels markers, increased lipid peroxidation, altered antioxidant enzyme activity impaired redox homeostasis. Treatment lithium other stabilisers may modulate which mechanism drug action. However, inconclusive data suggest need for further research clarify relationship Conclusions: plays an important pathophysiology offering directions Understanding complex interactions lead development more targeted therapies aimed at reducing damage improving patient health. The disorder and heterogeneity of low-molecular-weight amyloid-beta oligomers (AβOs) underlie their participation in multiple modes cellular dysfunction associated with the etiology Alzheimer's disease (AD). lack specified conformational states these species complicates efforts to select or design small molecules targeting discrete pathogenic states. Furthermore, AβOs alone may be therapeutically insufficient, as AD progresses a multifactorial, self-amplifying cascade. To address challenges, we have screened activity seven new candidates that serve Paramagnetic Amyloid Ligand (PAL) candidates. One candidate (HO-4450; PMT-307), six-membered ring attached ring, displays highest potency protecting against cell-derived Aβ. A preliminary low-dose evaluation model mice provides evidence modest treatment effects by HO-4450. results demonstrate retention native phenolic groups is advantageous hybrid Finally, remodeling secondary structures shows reasonable correlation between candidate's its ability reduce fraction antiparallel β-strand.
