Focused
ultrasound
(FUS)
is
an
emerging
noinvasive
technique
for
neuromodulation
in
the
central
nervous
system
(CNS).
To
evaluate
effects
of
FUS-induced
neuromodulation,
many
studies
used
behavioral
changes,
functional
magnetic
resonance
imaging
(fMRI)
or
electroencephalography
(EEG).
However,
readouts
are
often
not
easily
mapped
to
specific
brain
activity,
EEG
has
low
spatial
resolution
limited
surface
and
fMRI
requires
a
large
importable
scanner
that
limits
additional
manipulations.
In
this
context,
(fUSI)
holds
promise
directly
monitor
FUS
with
high
spatiotemporal
field
view,
comparatively
simple
flexible
setup.
fUSI
uses
ultrafast
Power
Doppler
Imaging
(PDI)
measure
changes
cerebral
blood
volume,
which
correlates
well
neuronal
activity
local
potentials.

The
PRISM
project,
funded
by
the
EU's
Innovative
Medicines
Initiative,
has
identified
a
transdiagnostic,
pathophysiological
relationship
between
integrity
of
default
mode
network
(DMN)
and
social
dysfunction.
To
explore
causal
link
DMN
behaviour,
we
employed
preclinical
back-translation
approach,
using
focal
demyelination
forceps
minor
to
disrupt
connectivity
in
mice.
By
applying
advanced
techniques
such
as
functional
ultrasound
imaging
automated
analysis
demonstrated
that
reduced
leads
impaired
interactions
increased
anxiety
Notably,
these
effects
were
reversible,
indicating
minor,
critical
fibre
tract
connecting
key
regions
prefrontal
cortex,
plays
crucial
role
function.
This
study
provides
direct
evidence
for
dysfunction,
with
potential
implications
developing
targeted
treatments
precision
psychiatry.

Abstract
Pharmacological
approaches
to
induce
N-methyl-D-aspartate
receptor
(NMDAR)
hypofunction
have
been
intensively
used
understand
the
aetiology
and
pathophysiology
of
schizophrenia.
Yet,
precise
cellular
molecular
mechanisms
that
relate
brain
network
dysfunction
remain
largely
unknown.
Here,
we
a
set
complementary
assess
functional
abnormalities
present
in
mice
underwent
7-day
subchronic
phencyclidine
(PCP
10mg/kg,
subcutaneously,
once
daily)
treatment.
Our
data
revealed
pharmacological
intervention
with
PCP
affected
cognitive
performance
auditory
evoked
gamma
oscillations
prefrontal
cortex
(PFC)
mimicking
endophenotypes
some
schizophrenia
patients.
We
further
assessed
PFC
function
identified
altered
neuronal
intrinsic
membrane
properties,
reduced
parvalbumin
(PV)
immunostaining
diminished
inhibition
onto
L5
pyramidal
cells.
A
decrease
strength
optogenetically-evoked
glutamatergic
current
at
ventral
hippocampus
(HPC)
synapse
was
also
demonstrated,
along
weaker
shunt
excitatory
transmission
by
local
interneurons.
On
macrocircuit
level,
ultrasound
measurements
indicated
compromised
connectivity
within
several
regions
particularly
involving
frontostriatal
circuits.
Herein,
reproduced
panel
induced
application

electrophysiological
signatures

findings

Abstract
The
