Sepsis can induce sepsis-associated encephalopathy (SAE), with Ulinastatin (UTI) serving a critical anti-inflammatory role. This study aimed to identify the hub genes in an SAE mouse model following UTI intervention and investigate underlying molecular mechanisms. Through differential expression analysis obtain differentially expressed (DEGs), ie, vs CLP (DEGs1) Con (DEGs2). After taking intersection of opposite trends these two parts immune-related (IRGs), DE-IRGs were obtained. Hub protein-protein interaction (PPI) network then determined using six algorithms from Cytohubba plugin Cytoscape. Gene set enrichment (GSEA) was employed explore functional relevance genes. Additionally, immune microenvironment across three groups compared, gene-related drugs predicted online database. Finally, qRT-PCR used validate hippocampal tissue mice. RNA sequencing obtained 864 (DEGs) (CLP Con) 279 DEGs (UTI CLP). Taking yielded 165 DEGs. Six key (ICAM - 1, IRF7, IL 1β, CCL2, 6 SOCS3) screened by algorithms. Immune infiltration found that Treg cells reversed after treatment diseased state. A total 106 gene related predicted, among which BINDARIT CCL2 LIFITEGRAST ICAM1 showed particularly high affinities. The qRT PCR verification results consistent results. In conclusion, ICAM-1, IL-1β, IL-6, SOCS3 identified as potential therapeutic targets mice treated UTI. offers theoretical support for option SAE.

Background/Objectives: Decompensated cirrhosis is characterized by systemic inflammation and innate adaptive immune dysfunction.
