Occupational allergic respiratory disease in healthcare workers due to occupational exposure has received widespread attention. At the same time, evidence of altered flora associated with development allergy been found relevant epidemiologic studies. It is concern that composition nasopharyngeal differs significantly from non-healthcare factors, a particularly high prevalence carriage pathogenic and drug-resistant bacteria. Diabetic nephropathy (DN) is one of the major causes end-stage renal disease. This study aimed to explore internal relationship between metabolic processes and autoimmune responses in patients with DN via untargeted metabolomics Olink proteomics. The serum 10 who were diagnosed healthy individuals Animal models used validate characterized genes. Correlation analysis differentially abundant metabolites expressed proteins revealed that SIRT2 might be a key hub linking energy metabolism innate immune responses. KEGG enrichment showed HIF-1 signaling pathway cell carcinoma co-enriched pathways inflammatory response. VEGFA plays vital role these two pathways. ability regulate expression has been demonstrated. In vivo experiments SIRT2, VEGFA, HIF-1α highly kidneys mice diabetic nephropathy. conclusion, our combines proteomics provide valuable insights into synergistic roles disorders DN. data suggest may target affecting processes.

The global prevalence of allergic rhinitis (AR) remains high, posing challenges due to its chronic nature and propensity for recurrence. Gut microbiota dysbiosis contributes immune dysregulation, impacting AR pathogenesis. Limosilactobacillus reuteri (L. reuteri) has great potential in regulating function alleviate symptoms. However, the specific active components mechanisms underlying therapeutic effects remain incompletely clarified. This study aimed explore L. metabolites alleviating AR. mouse model was constructed using ovalbumin (OVA). analysis hematoxylin-eosin staining (HE staining) enzyme-linked immunosorbent assay (ELISA) suggested that alleviated nasal inflammation, suppressed aberrant Th2 responses, modulated balance Treg Th17 cytokines. 16S rRNA sequencing untargeted metabolic revealed restored gut composition significantly increased abundance Ligilactobacillus metabolite luteolin (LO). Through ELISA Western blotting analysis, LO treatment Th1/Th2 Treg/Th17 cytokine MAPK/STAT3 signaling pathway mice. highlights as a key contributing anti-inflammatory reuteri, suggesting avenues future strategies management.

ABSTRACT Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) targets epithelial cells in the tract, triggering an proinflammatory response and chronic lung inflammation. Probiotic supplementation has shown promise reducing nasopharyngeal SARS-CoV-2 viral load, diminishing symptom frequency duration, mitigating inflammation; thus, it is a potential strategy for treating disease 2019 (COVID-19), which caused by infection. In this study, we evaluated effects of oral administration Lactobacillus plantarum GUANKE strain, gram-positive bacterium originally isolated from healthy individual, on infection human ACE2 transgenic mouse model. We found that significantly reduced inflammatory cell infiltration pulmonary interstitial exudation mice. The transcription CCL2 , TNFA IL1B IL6, IL17C lungs was reduced. protein levels TNF-α, IL-1β, IL-6, IL-17 were lower GUANKE-treated mice than control load saline-treated mice, although difference did not reach statistical significance. L. can decrease SARS-CoV-2-induced inflammation suggesting its use as agent IMPORTANCE Most otherwise individuals develop only mild or moderate symptoms (COVID-19) current strains severe (SARS-CoV-2), virus replication mainly confined to upper tract; however, infect tract promote been reduce overall number duration symptoms, attenuate clinical trials. showed novel strain alleviated pneumonia production cytokines suppressed, moderately load. become candidate drug treatment COVID-19 other infections.
