Abstract Background Adenoid cystic carcinoma (ACC) is a rare glandular malignancy, commonly originating in salivary glands of the head and neck. Given its protracted growth, ACC usually diagnosed advanced stage. Treatment limited to surgery and/or adjuvant radiotherapy, which often fails prevent disease recurrence, no FDA-approved targeted therapies are currently available. As such, identification new therapeutic targets specific crucial for improved patients’ outcomes. Methods After thoroughly evaluating gene expression signaling patterns characterizing ACC, we applied PandaOmics (an AI-driven software platform novel target discovery) on unique transcriptomic dataset 87 primary ACCs. Identifying protein arginine methyl transferase 5 (PRMT5) as putative candidate with top-scored druggability, next determined applicability PRMT5 inhibitors (PRT543 PRT811) using cell lines, organoids, patient derived xenograft (PDX) models. Adenoid cystic carcinoma (ACC) is a rare malignancy predominantly originating in the salivary glands, but capable of emerging other secretory-gland-rich anatomical sites, including reproductive tract, trachea, esophagus, and skin. Although characterized by its slow growth, ACC notably has concerning propensity for perineural spread often associated with late distant metastasis. Background/Objectives: Salivary adenoid cystic carcinoma (ACC) is a rare but aggressive neoplasm that predominantly arises from the salivary glands, accounting for significant proportion of gland cancers. The aim this literature review to illustrate current insights on ACC with regards related genes and molecular pathways by analyzing original research articles period 2015–2025. Methods: An electronic search was performed between January February 2025 identify all investigating its pathways. conducted using MEDLINE (National Library Medicine)-PubMed restrictions concerning date publication. Primary tracheal tumors are rare, constituting approximately 0.1-0.4% of malignant diseases. Squamous cell carcinoma (SCC) and adenoid cystic (ACC) account for about two-thirds these tumors. Abstract Background adenoid cystic carcinoma (ACC), a rare malignancy often treated through multidisciplinary approach, is noted for its slow progression and aggressive behavior. This study focuses on the impact of histone modification drugs (HMD) ACC tumors implications treatment. Methods: by analyzing effects HMD Cancer Stem Cells (CSC) non-CSC tumor cells, we investigated inhibitory effect various histone-modifying compounds cells.
