ABSTRACT Outer membrane protein A (OmpA) plays a vital role in the interactions between Acinetobacter baumannii and host cells. Autophagy is defense mechanism that hinders intracellular replication of bacteria, thereby safeguarding cells against microbial infections. While it has been observed A. triggers cellular autophagy, precise its virulence OmpA this process remains uncertain. In study, we investigated effects (AbOmpA) on autophagy explored underlying molecular mechanisms. We found AbOmpA exerted autophagy-suppressive effect through inhibition CaMKK2 phosphorylation. Compared to wild-type strain, ompA -deletion mutant strain displayed considerably enhanced induction, via AMPK-ULK1 pathway. hindered starvation-induced while -Omp33 (AbOmp33) Escherichia coli -OmpA (EcOmpA) did not. Importantly, confirmed exogenous suppressed CaMKK2-AMPK-ULK1 pathway during infection. These findings reveal novel for AbOmpA-mediated evasion, providing new insights into pathogenesis IMPORTANCE significant clinical pathogen notorious causing infections hospitals. Its outer acts as factor helps bacteria evade defenses. bacteria. Our studies demonstrate inhibits process, signaling cascade, enhancing bacterial survival. This insight how bypasses sheds light infection's strategy suggests possible treatments.

Abstract Background Lung cancer is one of the most prevalent malignancies globally and leading cause cancer-related mortality. Although cisplatin a widely utilized chemotherapeutic agent, its clinical efficacy often hampered by significant toxicity undesirable side effects. Rosa canina, medicinal plant, has demonstrated range beneficial biological activities, including anti-inflammatory, anticancer, immunomodulatory, antioxidant, genoprotective Methods This study aimed to investigate potential canina enhance anticancer in dimethyl benz(a)anthracene-induced lung model using female rats. The animals were administered cisplatin, or combination both treatments. expression levels critical signaling molecules evaluated, phosphoinositide-3-kinase (PI3K), Akt, mammalian target rapamycin (mTOR), cleaved poly (ADP-ribose) polymerase (PARP-1), myeloid differentiation factor 88 (MyD88), tumor necrosis receptor-associated (TRAF), addition various autophagic markers. Furthermore, we assessed toll-like receptor 2 (TLR2), nuclear kappa B (NF-κB), apoptotic markers tissue, complemented histopathological examinations. Results combined treatment extract significantly inhibited cell proliferation downregulating PARP-1 TLR2/MyD88/TRAF6/NF-κB pathway, as well PI3K/Akt/mTOR pathway. Moreover, this therapy promoted autophagy apoptosis, evidenced elevated Conclusion Overall, findings suggest that enhances effects inhibiting while simultaneously inducing apoptosis. Thus, can be used adjuvant chemotherapy overcome limitations which may considered new approach during strategy. Graphical

Lung cancer (LC) is the leading cause of death worldwide. LC can be classified into small-cell lung (SCLC) and non-small-cell (NSCLC), with last subtype accounting for approximately 85% all diagnosed cases. Despite existence different types treatment this disease, development resistance to therapies tumor recurrence in patients have maintained need find new therapeutic options combat pathology, where natural products stand out as an attractive source search. Nordihydroguaiaretic acid (NDGA) main metabolite extracted from Larrea tridentata plant has been shown biological activities, including anticancer activity. In study, H1975, H1299, A549 cell lines were treated NDGA, its effect on viability, proliferation, metabolism was evaluated using a resazurin reduction assay, incorporation BrdU, ki-67 gene expression glucose uptake measurement, respectively. addition, combination NDGA clinical chemotherapeutics investigated MTT assay Combenefit software (version 2.02). The results showed that decreases viability proliferation NSCLC cells differentially modulates genes associated metabolic pathways. For example, LDH decreased analyzed. However, GLUT3 increased after 24 h treatment.
