Patients diagnosed with non-small cell lung cancer (NSCLC) usually have a poor prognosis, so it is critical to identify effective biomarkers for prognosis prediction. The aim of this study establish nomogram evaluate the prognostic significance blood markers in patients NSCLC and provide reference clinical work. A total 486 who were admitted hospital from January 2009 December 2019 retrospectively analyzed. cohort was divided into training set (n=340) validation (n=146). Eleven indicators selected as parameters by least absolute shrinkage selection operator (LASSO) model tumor marker inflammatory nutrition (TMIN) score. Univariate multivariate regression analyses performed TMIN-nomogram predicting overall survival (OS) progression-free (PFS). Receiver operating characteristic (ROC) curve, calibration curve decision analysis (DCA) used predictive performance model. TMIN score constructed 11 most valuable variables, including white cells (WBCs), neutrophils (N), platelets (PLT), albumin (ALB), globulin (GLB), prealbumin (PAB), carcinoembryonic antigen (CEA), cytokeratin fragment 21-1 (CYFRA21-1), fibrinogen (FIB), platelet/lymphocyte ratio (PLR), lymphocyte/monocyte (LMR), low-risk high-risk groups using optimal cutovers. showed good value both OS PFS. In addition, sex, smoke, pathological classification, American Joint Committee on Cancer stage (AJCC stage), diameter Eastern Cooperative Oncology Group-performance status (ECOG-PS) other strong correlation. confirmed that an independent risk factor PFS patients. Cancer remains a leading cause of mortality globally and major health burden, with chemotherapy often serving as the primary therapeutic option for patients advanced-stage disease, partially compensating limitations non-curative treatments. However, emergence resistance significantly limits its efficacy, posing clinical challenge. Moreover, heterogeneity mechanisms across cancer types complicates development universally effective diagnostic approaches. Understanding molecular chemoresistance identifying strategies to overcome it are current research focal points. This review provides comprehensive analysis key underlying resistance, including drug efflux, enhanced DNA damage repair (DDR), apoptosis evasion, epigenetic modifications, altered intracellular metabolism, role stem cells (CSCs). We also examine specific causes in highlight various targets involved resistance. Finally, we discuss aiming at overcoming such combination therapies, targeted treatments, novel delivery systems, while proposing future directions this evolving field. By addressing these barriers, lays foundation more therapies aimed mitigating

Manzamine A (MA), a bioactive compound derived from the marine sponge Haliclona sp., shows considerable therapeutic potential, particularly in treatment of various cancer types. Extracted with acetone and purified through chromatography, MA exhibits bioavailability 20.6% when administered orally rats, underscoring its feasibility for use. This disrupts key cellular mechanisms essential progression, including microtubule dynamics DNA replication enzymes, demonstrating strong anti-proliferative effects against multiple cell lines while sparing normal cells. Additionally, network pharmacology molecular docking studies reveal MA's interactions important targets related to lung such as EGFR SRC, bolstering potential novel anti-lung agent. Pathway analyses further indicate that influences critical signaling pathways involved tumor growth metastasis. Given urgent need effective treatments drug-resistant cancers limited toxicity profile MA, exploration pharmacological benefits mechanism could pave way new strategies cancer.

The current study aims to investigate the practice of bone metastasis management in patients with non-small cell lung cancer. An online questionnaire was administered 92 oncologists. A survey developed and revised by dedicated experts composed five sections: i) general work characteristics, ii) diagnostic issues, bone-targeted agents iii) radiotherapy iv) supportive care issues. Descriptive statistics applied. 18F-FDG PET is preferred evaluation for skeletal assessment both (62 %) without (54 lesions at CT scan; MRI (63 (61 are most chosen radiographic assessments when a oligoprogression suspected. number metastatic main factor considered deciding whether start (57 %). In choosing between agents, renal toxicity Over half participants did not stop systemic treatment during stereotactic (68 re-irradiation on progressive metastases least 6 months after prior (55 Overall, 64 % 41 assessed patients' body weight physical activity, respectively. Oral nutritional supplements or specific diet were recommended 34 46 clinicians; 40 them also advised their increase activity levels, while 54 worried that exercise might risk skeletal-related adverse events. Lung-cancer clinicians pay great attention metastases-related diagnostic, whereas integration approaches seem less standardized.

Abstract Background Osteosarcoma has been a common bone malignancy occurring in children and adolescents. Attributing to high tumor heterogeneity, none specific breakthrough received targeted gene therapy for osteosarcoma, although it’s still of great potential immunotherapy clinical application. In the study, 5 GEO profiles containing transcriptome information 109 osteosarcoma samples, single cell sequencing data composed 6 cases as well 43 local hospital tissue samples were combine used identify promising immune related candidate genes osteosarcoma. Methods Based on microarrays from database profile IMMPORT database, differently expressed meanwhile candidates comparing normal control identified. Then, protein-protein interaction network (PPI), survival analysis followed by LASSO succession applied construct signature based selected genes. After understanding basic genetic physicochemical properties evaluating prognosis risk association using cancer its with microenvironment features including macrophages included various cells infiltration, different checkpoints expression, signaling pathways involvement next step assessed. Results From datasets which contains total 108 level PPI highlighted cluster Further, ROC curve Cox regression assisted scaled hub down 2 key genes, namely STC2 FPR1, was constructed them. double staining immunochemistry (IHC) experiment date revealed that mainly cells, meanwhile, FPR1 mostly enriched focused also main type microenvironment. Moreover, combining STC2/FPR1 dual associated distribution multiple checkpoints, especially TIM-3. correlation between other death (ICD) ESTIMATE score additionally evaluated. Conclusions analysis, constructed. Immune indicated infiltration ans it TIM-3 expression. The results shall benefit further researches assist revealing prediction markers immunotherapy.
