Alzheimer's disease (AD) is a common neurodegenerative disease. In AD-associated neuroinflammation, astrocytes play key role, finding glial activation both in patients and animal models. The endocannabinoid system (ECS) neurolipid signaling with anti-inflammatory neuroprotective properties implicated AD. Astrocytes respond to external cannabinoid signals also have their own signaling. Our main objective describe the machinery present hippocampal from 3×Tg-AD mice determine if they are actively involved process. Primary cultures of hippocampus non-Tg offspring were carried out. We analyzed gene expression astrogliosis markers, components ECS Ca2+ show low inflammatory activity (Il1b, Il6, Gls) flow (P2rx5 Mcu), associated (Cnr1 Cnr2). Despite decades of rigorous research and numerous clinical trials, Alzheimer’s disease (AD) stands as a notable healthcare challenge this century, with effective therapeutic solutions remaining elusive. Recently, the endocannabinoid system (ECS) has emerged an essential target due to its regulatory role in different physiological processes, such neuroprotection, modulation inflammation, synaptic plasticity. This aligns previous showing that cannabinoid receptor ligands have potential trigger functional structure neuronal brain networks, potentially impacting memory processing. Therefore, our study aims assess effects prolonged, intermittent exposure (over 90 days) JWH-133 (0.2 mg/kg) EU-GMP certified Cannabis sativa L. (Cannabixir® Medium Flos, 2.5 on recognition memory, well their influence metabolism expanded APP/PS1 mice. Chronic therapy resulted reduced anxiety-like behavior partially reversed cognitive deficits. Additionally, reduction was observed both number size Aβ plaque deposits, along decreased cerebral glucose metabolism, decline expression mTOR CB2 receptors. Furthermore, revealed enlarged astrocytes enhanced M1 mAChR mice subjected treatment. Our findings highlight pivotal involvement extended pathological aspects associated AD, presenting preclinical evidence support continued exploration assessment for AD

The findings concerning the association between endocannabinoid system (ECS) and Alzheimer's disease (AD) exhibited inconsistencies when examining expression levels of endocannabinoids. This study aimed to provide a comprehensive summary studies regarding alterations ECS in AD. Six databases were thoroughly searched for literature select relevant investigating AD, including changes cannabinoid receptors (CB1R CB2R), endocannabinoids (2-AG AEA), their associated enzymes (FAAH MAGL). Traditional meta-analysis evaluated results showed no significant differences components healthy controls AD patients. However, subgroup analysis revealed significantly lower CB1R than controls, particularly using western blot (SMD = -0.88, p < 0.01) testing frontal cortex -1.09, 0.01). For HPLC, indicated higher 2-AG 0.46, 0.02). Network examined rank compared that MAGL largest increase decrease relative control group. Based on traditional network meta-analysis, we proposed patients may present decreased increased its degrading enzyme MAGL. Our contribute growing body research supporting therapeutic potential modulation management
