To determine the efficacy and safety of Yinmei Kuijie decoction combined with 5-aminosalicylic acid (5-ASA) in treating mildly to moderately active ulcerative colitis (UC) under real-world conditions. This multicenter, prospective, non-randomized, observational study will be conducted settings. A total 204 eligible patients consecutively enrolled study. Patients combination treatment group receive 5-ASA, whereas those control treated 5-ASA alone. The primary endpoint a clinical response at week 12, defined as ≥3 point ≥30% reduction from baseline Mayo score ≥1 rectal bleeding or = 0 1. Secondary endpoints 12 include health-related quality life, mucosal healing, inflammation indicators. results this may provide evidence UC principles. basis for further confirmatory studies on decoction.

Colorectal cancer (CRC) is a significant global health burden, with high morbidity and mortality rates. It often diagnosed at middle to advanced stage, affecting approximately 35% of patients the time diagnosis. Currently, chemotherapy has been used improve patient prognosis increase overall survival. However, can also have cytotoxic effects lead adverse reactions, such as inhibiting bone marrow hematopoiesis, causing digestive dysfunction, hand-foot syndrome, even life-threatening conditions. In response these effects, researchers proposed using Traditional Chinese Medicine (TCM) an option treat cancer. TCM research focuses on prescriptions, herbs, components, which form essential components current in medicine. The study implementation prescriptions herbs demonstrate its distinctive holistic approach therapy, characterized by applying multi-component multi-target treatment. TMC advantages developing new drugs they consist single ingredients, require smaller medication dosages, precise measure pharmacodynamic clear mechanism action compared herbs. further still needed determine whether fully substitute therapeutic efficacy prescriptions. This paper presents comprehensive analysis advancements made components. findings this serve theoretical basis for who are interested exploring potential treatment colorectal

Colorectal cancer (CRC) is the most prevalent globally, and its traditional treatment modalities commonly encompass radiation therapy, chemotherapy, surgery administration of cytotoxic drugs. Currently, novel chemotherapy drugs that combine Chinese medicine (TCM) with herbal extracts exhibit superior comprehensive benefits. Herein, we delved into an article authored by Wang et al , focusing specifically on pharmacological effects "Herba Patriniae Coix seed (HC)" their targeted mechanisms in combating CRC. From perspective TCM philosophy, damp-heat stagnation toxicity are cardinal pathogenic factors underlying HC, renowned for abilities to antipyretic enhance diuresis, have demonstrated promising efficacy preliminary studies These findings offer potential insights favor fostering anti-cancer medications.

Astragali Radix (AR) - Curcumae Rhizoma (vinegar processed, CR) herb pair was recorded in 'YIXUE ZHONGZHONG CANXILU' to treat colitis-associated colorectal cancer (CAC). Hedysari (HR) categorized under the AR entry 'SHENNONG BENCAO JING'. HR is still an alternative paired with CR clinically northwest China. Praeparata Cum Melle (HRPCM) a product that fries honey enhance therapeutic effect. However, mechanism of HRPCM (HRCR) CAC needs be further elucidated. HRCR-MIAS were prepared using eversion intestinal sac method. UHPLC Q-Exactive-MS investigated compositions HRCR-MIAS. Then, HRCR mice predicted based on network pharmacology analysis combination The pharmacodynamic effects for SW620 colon cells invested vitro. efficacies low-, middle-, and high-dose groups (HRCR-L, 3.413 g/kg; HRCR-M, 6.825 HRCR-H, 13.650 g/kg) explored. Enzyme-linked immunosorbent assay (ELISA) kits employed inflammatory factors levels, like IL-1β, IL-6, IL-10, TNF-α serum. expressions permeability proteins, such as Claudin-1, Occludin, ZO-1, detected via immunohistochemical (IHC) analysis. Finally, signalling verified by Western blot (WB). 855 common components identified HRCR-MIAS, 25 specific pointed out. Hedysari Radix Praeparata Cum Melle (HR) has a potential to decrease the risk of chronic colorectal inflammation developing cancer. Our study found that HR attenuated pathological state, improved survival rate, diminished K-Ras, IL-1β, IL-6 and TNF-α levels, increased levels intestinal barrier function proteins including claudin-1, E-cadherin mucin-2 in colitis-associated cancer (CAC) mice. Moreover, maintains flora homeostasis fecal metabolic profiling through balancing abundances bacteroides prevotellaceae_UCG-001, staphylococcus, turicibacter, rikenella further regulates galactose metabolism. The spearman correlation analysis showed proteins, gut microbiota differential metabolism were significantly correlated with each other. This confirmed mechanism alleviates CAC by diminishing symptoms inflammation, thereby improving preserving profiling.

BACKGROUND Herba Patriniae and Coix seed (HC) constitute a widely utilized drug combination in the clinical management of colorectal cancer (CRC) that is known for its diuretic, anti-inflammatory, swelling-reducing properties. Although efficacy has been demonstrated setting, active compounds their mechanisms action CRC treatment remain to be fully elucidated. AIM To identify active, CRC-targeting components HC elucidate involved. METHODS Active were identified screened using databases. Targets each component predicted. CRC-related targets obtained from human gene Interaction between identified. A “drug-ingredient-target” network was created core Gene Ontology (GO) Kyoto Encyclopedia Genes Genomes (KEGG) analyses conducted key pathways Molecular docking executed. In vitro experiments validated monomers. RESULTS Nineteen identified, with acacetin as primary compound. The predictive analysis 454 HC. Intersection mapping 2685 yielded 171 intervention targets, including 30 targets. GO KEGG indicated may influence phosphoinositide 3-kinase (PI3K)/Akt signaling pathway. showed exhibited an optimal interaction AKT1 , identifying PI3K AKT P53 genes likely targeted by during treatment. Acacetin inhibited HT-29 cell proliferation migration, well promoted apoptosis, . Western blotting revealed increased p53 cleaved caspase-3 expression decreased levels p-PI3K p-Akt survivin, which contributed apoptosis. CONCLUSION Acacetin, principal compound pair, migration cells apoptosis through PI3K/Akt/p53
