ABSTRACT In traditional Chinese medicine, Ramulus mori (Sangzhi) is used to lower blood sugar and treat diseases such as diabetes, its therapeutic effects are attributed an abundance of bioactive compounds. This study employed a comparative analysis from three geographical regions within China. The samples included Morus atropurpurea Roxb Guangdong (GDS), australis Poir Jiangsu (JS), nigra Linn, which native Xinjiang (XJYS). discovered significant differences in total polyphenol flavonoid content among various (Sangzhi), with those XJYS showing the highest levels. Utilizing UPLC‐ESI‐MS/MS technology, 128 phenolic metabolites were identified, 17 differential screened. Comparison GDS, JS, exhibited 12 10 different metabolites, respectively, 8 observed between JS XJYS. Furthermore, higher than other two varieties. Additionally, some upregulated have been confirmed possess variety pharmacological health benefits. Kyoto Encyclopedia Genes Genomes linked these biosynthesis, qRT‐PCR corresponding gene expression patterns. These findings provided theoretical basis for use pharmaceutical food industries.

Acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) are enzymes that break down reduce the level of neurotransmitter acetylcholine (ACh). This can cause a variety cognitive neurological problems, including Alzheimer's disease. Taxifolin is natural phytochemical generally found in yew tree bark has significant pharmacological properties, such as being anti-cancer, anti-inflammatory, antioxidant. The binding affinity inhibitory potency taxifolin to these were evaluated through molecular docking dynamics simulations followed by MMPBSA approach, results significant. Taxifolin's for AChE-taxifolin complex was -8.85 kcal/mol, with an inhibition constant 326.70 nM. It observed interact hydrogen bonds. In contrast, BChE-taxifolin energy be -7.42 it significantly nearly equal standard inhibitor donepezil. simulation signified interactions studied enzymes. total free -24.34 while -16.14 kcal/mol. present research suggests strong ability bind inhibit AChE BChE could used manage neuron-associated problems; however, further required explore taxifolin's therapeutic potential using animal models

Cases of iatrogenic cerebral amyloid angiopathy (CAA) have been increasingly reported recently, particularly those associated with neurosurgery. Preclinical studies shown taxifolin to be promising for treating CAA. We describe a young 42-year-old man history childhood traumatic brain injury that required craniotomy hematoma evacuation. He later presented recurrent lobar intracerebral hemorrhage (ICH) decades later, which was histologically confirmed Serial 11 C-Pittsburgh compound B positron emission tomography ( C-PiB-PET) imaging showed 24% decrease in global standardized uptake value ratio (SUVR) at 10 months after use. During this period, the patient experienced clinical improvement improved consciousness and reduced ICH frequency, may partly attributable potential amyloid-β (Aβ) clearing effect taxifolin. However, seemed diminished 15 months, CAA should considered patients presenting neurosurgery, serial C-PiB-PET scans warrant further validation as strategy monitoring treatment response candidate Aβ-clearing therapeutic agents such

Introduction. The structure of dihydroquercetin (DHQ) is characterized by two chiral centers at positions 2 and 3 the benzopyran cycle, resulting in possible diastereomers: trans - cis -isomers. Therefore, development methods for qualitative quantitative control DHQ diastereomers analyzed samples essential patient safety management. Nuclear magnetic resonance (NMR) spectroscopy one physicochemical that can be used this purpose. Aim. study objective was to accumulate analytical structural characteristics -DHQ NMR spheroidal form flavonoid (DHQs). Materials Methods. 1D 1 H, H-COSY, H-NOESY, 13 C-HSQC spectra were acquired 298 K on an 800 MHz spectrometer equipped with a TXI triple probe. number scans 32. mixing time NOESY experiment 400 ms. H C using CcpNmr software. dihedral angles calculated applying Karplus equation. Results discussion. In -DHQ, chemical shift values H2 H3 are 4.93 ppm 4.52 ppm, respectively, they 5.31 4.20 respectively. spin-spin coupling constants between 12.00 Hz 2.40 Hz, Thus, -isomers 154° 64°, We found DHQs contains 12.5 % -isomer. Conclusion. Our experiments confirmed discriminate based cross-peaks H3. second major finding method considered as more selective analysis than HPLC UV detection without reference. One most important results drug updated information parameters liquid phase.

Aromadendrin and taxifolin are two flavanonols (derived from the precursor naringenin) displaying diverse beneficial properties for humans. The carbon skeleton of these flavonoids may be transformed by human gastrointestinal microbiota into other compounds, like auronols, which exert different interesting biological activities. While research in has become a certainly extensive field, studies about auronols still scarce. In this work, versions key plant enzyme biosynthesis, flavanone 3-hydroxylase (F3H), been screened selecting best one de novo production compounds bacterial factory Streptomyces albidoflavus UO-FLAV-004-NAR, naringenin overproducer strain. This screening rendered 2.6 μg/L aromadendrin 2.1 mg/L final titers. Finally, expression chalcone isomerase (CHI) gut bacterium Eubacterium ramulus direct conversion (after feeding experiments) 38.1% (+)-aromadendrin maesopsin 74.6% (+)-taxifolin alphitonin. Moreover, heterologous biosynthesis 1.9 alphitonin was accomplished means co-culture strategy producer S. CHI-expressing Escherichia coli, after observation high instability culture medium. study addresses significance time optimization selection appropriate enzymes depending on desired product. To our knowledge, is first that accomplished.
