Berberine (BBR) is a compound derived from Chinese herbal medicine, known for its anticancer properties through multiple signaling pathways. However, whether BBR can inhibit tumor growth by participating in ferroptosis remains unconfirmed. In this study, we demonstrated that berberine synergistically inhibited NSCLC combination with inducers, and down-regulated the mRNA protein expression of SLC7A11, GPX4, NRF2, resulting accompanied significant depletion GSH, aberrant accumulation reactive oxygen species malondialdehyde. lung cancer allograft model, treatment exhibited enhanced effects compared to using either drug alone. Notably, p53 critical determining sensitivity. We found did not elicit synergistic effect cells mutation or exogenous mutant p53. These findings provide insight into mechanism which induces regulatory role process. It may guide development new strategies treating NSCLC, offering great medical potential personal diagnosis treatment.

Oxidative stress is a key factor in the development of chronic diseases such as type 2 diabetes, cardiovascular diseases, and liver disorders. Antioxidant therapies that target oxidative damage show significant promise preventing treating these conditions. Berberine, an alkaloid derived from various plants Berberidaceae family, enhances cellular defenses against through several mechanisms. It activates AMP-activated protein kinase (AMPK) pathway, which reduces mitochondrial reactive oxygen species (ROS) production improves energy metabolism. Furthermore, it boosts activity antioxidant enzymes like superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), thus protecting cells damage. Abstract Atherosclerotic cardiovascular disease remains a preeminent cause of morbidity and mortality globally. The onset atherosclerosis underpins the emergence ischemic diseases, including coronary heart (CHD). Its pathogenesis entails multiple factors such as inflammation, oxidative stress, apoptosis, vascular endothelial damage, foam cell formation, platelet activation. Furthermore, it triggers activation diverse signaling pathways Phosphatidylinositol 3‐kinase/protein kinase B (PI3K/Akt), NF‐E2‐related factor 2/antioxidant response element (Nrf2/ARE), Notch pathway, peroxisome proliferator‐activated receptor (PPAR), nucleotide oligo‐structural domain‐like thermoprotein structural domain‐associated protein 3 (NLRP3), silencing information regulator 2‐associated enzyme 1 (Sirt1), nuclear transcription factor‐κB (NF‐κB), Circular RNA (Circ RNA), MicroRNA (mi Transforming growth factor‐β (TGF‐β), Janus kinase‐signal transducer activator (JAK/STAT). Over recent decades, therapeutic approaches for have been dominated by utilization high‐intensity statins to reduce lipid levels, despite significant adverse effects. Consequently, there is growing interest in development safer more efficacious drugs modalities. Traditional Chinese medicine (TCM) offers vital strategy prevention treatment diseases.
