Introduction Osteosarcoma, a malignant bone tumor, poses significant treatment challenges, necessitating the development of alternative therapeutic strategies. Aerva lanata (A. lanata), medicinal plant with traditional use in various healthcare systems, has anti-cancer properties. This study looks at oncolytic effect A. extract on osteosarcoma cell lines (sarcoma osteogenic-Saos2). Aim The aim this was to investigate Saos2 through apoptotic signaling pathway. Materials and methods prepared using Soxhlet extraction, its cytotoxic effects cells were assessed 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Real-time polymerase chain reaction (RT-PCR) analysis gene activity used assess extract's pathways. Results MTT assay demonstrated dose-dependent decrease proliferation following concentrations ranging from 50 μg 200 μg. standard deviations observed ranged 1.414 7.071. Gene expression revealed that led reduction messenger ribonucleic acid (mRNA) levels anti-apoptotic marker B-cell lymphoma 2 (Bcl2), 1 0.535. Conversely, it induced an increase mRNA tumor suppressor protein p53, 1.835. These findings suggest modulates pathways Bcl2 p53 genes. Conclusion exhibits promising against lines, inducing apoptosis by downregulating increasing p53. study's may be useful as natural for osteosarcoma.
