This study offers promising insights into the anti-obesity potential of Pleurotus ferulae, an edible mushroom valued in Asian cuisine for its nutritional benefits. A hot water extract P. ferulae (PWE) administered to high-fat diet-induced obese mice over 8-week period significantly reduced their body weight gain and fat accumulation. PWE not only improved metrics but also positively influenced serum lipid profile by lowering total cholesterol low-density lipoprotein levels. In vitro studies using 3T3-L1 adipocytes showed that inhibited adipocyte differentiation accumulation downregulating key adipogenic transcription factors, particularly PPARγ C/EBPα, as well related lipogenic genes involved synthesis storage, such Fabp4, Fasn, Scd1. Chemical analysis revealed is rich polysaccharides, which have been associated with various health benefits, including anti-obesity, anti-diabetic, anti-cancer properties. These findings suggest bioactive compounds may serve functional food components could potentially be applied prevention management obesity other metabolic disorders.

Golden Retrievers have a high risk of obesity, which is prevalent in dogs and associated with inflammation cancer, impairing the health life expectancy companion animals. Microbial metabolite biomarkers been proposed for identifying presence obesity humans rodents. However, effects on microbiome metabolome remains unknown. Therefore, this study was designed to evaluate signatures serum biochemistry indexes, gut microbiota plasma metabolites non-obese obese Retrievers, aiming recognize potential canine obesity. A total 8 (Ctrl group) (Obe were included present collect blood feces samples measurements. The carnitine palmitoyltransferases (CPTs) play a key role in controlling the oxidation of long-chain fatty acids and are potential therapeutic targets for diseases with strong metabolic component, such as obesity, diabetes, cancer. Four distinct proteins CPT1A, CPT1B, CPT1C, CPT2, differing tissue expression catalytic activity. CPT1s finely regulated by malonyl-CoA, metabolite whose intracellular levels reflect cell's nutritional state. Although CPT1C does not exhibit significant activity, it is capable modulating functioning other neuronal proteins. Structurally, all CPTs share Y-shaped tunnel that allows entry 2 substrates accommodation acyl group hydrophobic pocket. Several molecules targeting these enzymes have been described, some showing normalizing blood glucose diabetic patients, others that, through central mechanism, anorexigenic enhance energy expenditure. However, given critical roles certain tissues, heart, liver, brain, essential to fully understand differences between various isoforms. We analyze detail structure proteins, their cellular physiological functions, also describe drugs identified date having inhibitory or activating capabilities This knowledge will support design new specific each isoform, development nanomedicines can selectively target particular tissues cells. SIGNIFICANCE STATEMENT: Carnitine palmitoyltransferase (CPT) gatekeepers acid oxidation, great pharmacological treat like In recent years, progress has made understanding 3-dimensional pathophysiological functions. A deeper CPT family members enable selective approaches fewer side effects.

Background: Obesity is a growing global health concern, often accompanied by dyslipidemia, contributing to cardiovascular risk. Understanding the patterns of dyslipidemia in different glycemic states crucial for targeted interventions. This study compares normoglycemic and prediabetic obesity improve clinical management strategies. Methods: The analyzed complete lipid profiles 138 subjects, comparing medians, prevalence, diagnostic performance, risk assessment each parameter across 54 non-obese (NO), 44 obese (NG-OB), 40 pre-diabetic (PreDM-OB) groups. Results: Elevated total cholesterol (TC) low-density lipoprotein (LDL) were most prevalent forms observed (45.35% 43.53%, respectively). Stratification status revealed that triglyceride (TG) levels elevated both NG-OB PreDM-OB groups, with more marked increase latter group (73.07 mg/dL vs. 97.87 121.8 mg/dL, LDL showed better performance higher odds ratios (OR) (AUC = 0.660, p 0.006; OR 2.78, 0.022). Conversely, low high-density (HDL) was common exhibited significant values 0.687, 0.002; 3.69, 0.018). Importantly, all obesity, TC/HDL showing highest predictive ability prediabetes 0.7491, < 0.001). Conclusions: These findings unique abnormalities obesity. Future research should explore effects on obesity-associated complications.
