Torpor is a state used by several mammals to survive harsh winters and avoid predation, characterized drastic reduction in metabolic rate followed decrease body temperature, heart rate, many physiological variables. During torpor, all organs systems must adapt the new low-energy expenditure conditions preserve homeostasis. These adaptations may be exploited translational perspective fields. Recently, features of torpor were shown mimicked non-hibernators inhibition neurons within brainstem region Raphe Pallidus. The resemblance this artificial state, called synthetic with natural has so far been described only terms, but no data have regarding induced morphological changes. Here, we show first description ultrastructural changes liver, kidney, lung, skeletal muscle, testis 6-hours Pallidus non-hibernating species, rat.

Abstract Hibernating brown bears, due to a drastic reduction in metabolic rate, show only moderate muscle wasting. Here, we evaluate if ATPase activity of resting skeletal myosin can contribute this energy sparing. By analyzing single fibers taken from the same either during hibernation or summer, find that hibernating bears have mild decline force production and significant activity. Single fiber proteomics, western blotting, immunohistochemical analyses reveal major remodeling mitochondrial proteome hibernation. Furthermore, using bioinformatical approaches blotting phosphorylated light chain, known stimulator basal activity, is decreased disused muscles. These results suggest limits loss by reducing indicating possible role for modulation multiple wasting conditions.

Abstract The genus Myotis is one of the largest clades bats, and exhibits some most extreme variation in lifespans among mammals alongside unique adaptations to viral tolerance immune defense. To study evolution longevity-associated traits infectious disease, we generated near-complete genome assemblies cell lines for 8 closely related species . Using genome-wide screens positive selection, analyses structural variation, functional experiments primary lines, identify new patterns adaptation contributing longevity, cancer resistance, interactions bats. We find that bats have significant risk across demonstrate a DNA damage response cells long-lived M. lucifugus also evidence abundant viruses - but not RNA other sharp contrast with mammals, potentially role as reservoirs zoonoses. Together, our results how genomics derived from diverse taxa uncover molecular bases non-model organisms.

Abstract Hibernation is a fascinating physiological phenomenon that dramatically reduces basal metabolism and thermogenesis, resulting in large deviation body temperature (Tb) from homeothermic ranges mammals. Although high-resolution long-term Tb recording wild or laboratory animals has become possible through data loggers, few standardized methods to analyze details of hibernation patterns are available, making it difficult reproduce compare the results across different studies species. To facilitate analysis accelerate research, we developed an open-source program, tools measurement interpretation (TOHMIN). As proof concept, analyzed dataset two pilot on (1) effects distinct diets (2) differences between males females mammalian hibernator, Syrian hamster ( Mesocricetus auratus ), found previously undetectable fine-scale patterns. First, types affected duration periodic arousal. Second, maintained higher temperatures during arousal than males. Third, pre-hibernation period was negatively correlated with for this Thus, TOHMIN accelerates assess various experimental manipulations phenotypes
