Abstract Land plants produce a cuticle, an extracellular hydrophobic layer that covers aerial organs and is involved in many critical protective roles, most notably preventing desiccation. The predominant component of the cuticle lipidic polyester, cutin, which deposited epidermal primary cell wall. Most cutin tomato fruit, model for research, polymerized by GDSL-hydrolase enzyme CUTIN SYNTHASE-LIKE 1 (CUS1). However, other enzymes assembly remain to be identified characterized. In this current study, we investigated whether GDSL-hydrolases are highly expressed fruit epidermis might also contribute polymerization restructuring. Candidates include homologs Arabidopsis thaliana CUTICLE DESTRUCTIVE FACTOR (CDEF1), has been reported catalyze hydrolysis, as well phylogenetically diverse distantly related GDSL-hydrolases. We determined members CUS CDEF families can transesterification precursors vitro , modify structure semi- vivo assays. GDSL esterase/lipases (GELPs) play important roles in plant growth, development, and response to biotic abiotic stresses. Presently, an extensive in-depth analysis of GELP family genes cotton is still not clear enough, which greatly limits the further understanding function regulatory mechanism.A total 389 were identified three species Gossypium hirsutum (193), G. arboreum (97), raimondii (99). These GELPs could be classified into groups eight subgroups, with same group have similar gene structures conserved motifs. Evolutionary event showed that tend diversified at spatial dimension certain conservative time dimension, a trend potential continuous expansion future. The orthologous or paralogous among different genomes/subgenomes indicated inheritance from genome-wide duplication during polyploidization, derived chromosomal segment tandem replication. A/D subgenome underwent least large-scale replication events evolutionary process period 0.6-3.2 MYA, two between 0.6-1.8 MYA associated tetraploidization, 2.6-9.1 occurred diploidization. diverse expression patterns tissue ovule fiber stresses, combining existing cis-elements promoter regions, suggesting involvements functions diversification mechanisms hormone-mediated manner.Our results provide systematic comprehensive mechanism family, offer effective reference for genetic improvement utilization GELPs.

Wheat is the most widely grown crop in world; its production severely disrupted by increasing water deficit. Plant roots play a crucial role uptake of and perception transduction deficit signals. In past decade, mechanisms drought tolerance have been frequently reported; however, transcriptome metabolome regulatory network root responses to stress has not fully understood wheat. this study, global transcriptomic metabolomics profiles were employed investigate responding stresses using drought-tolerant (DT) drought-susceptible (DS) wheat genotypes. The results showed that compared with control group, exposed polyethylene glycol (PEG) had 25941 differentially expressed genes (DEGs) more upregulated found DT (8610) than DS (7141). Gene Ontology (GO) Kyoto Encyclopedia Genes Genomes (KEGG) analysis DEGs genotype preferably enriched flavonoid biosynthetic process, anthocyanin biosynthesis suberin biosynthesis. integrated DT, KEGG pathways, including arginine proline metabolism, shared accumulated metabolites (DAMs) at 6 h after treatment (HAT) pathways alanine, aspartate, glutamate metabolism carbon 48 HAT, while DS, DAMs only included HAT amino acids HAT. Yellow pigment content, mainly due to the accumulation of carotenoids, is a quality trait in durum wheat grain as it confers bright yellow color pasta preferred by consumers. Also, carotenoids are essential nutrients exerting important biological functions human health. Consequently, biofortification strategies have been developed many crops increase carotenoid content. In this context, esterification emerging new breeding target for biofortification, esters found promote both and stability. Until recently, no identified significant proportions grains, interspecific programs started transfer ability from common Hordeum chilense.In work, XAT-7A1 gene responsible wheat. Sequencing, copy number variation mapping results show that organized tandem or proximal GDSL esterase/lipase copies chromosome 7A. Three haplotypes described: Type 1 copies, associated with high levels (diesters monoesters) production expression development; 2 present landraces low (monoesters) esters; 3 without signal peptide, resulting zero-ester phenotypes.The identification necessary step make available bread breeding, which should be focused on haplotype, may assessed single since inherited together.
