Background: Escherichia coli (E. coli) is a common opportunistic bacterial pathogen in both human and animal populations. Fatty acids serve as the central carbon energy source, process mediated by fatty acid-coenzyme A (CoA) ligases encoded fad genes such FadK. However, function mechanism of FadK remain unclear. Methods: The three-dimensional structure was modeled using AlphaFold2. After expression purification, monomeric successfully isolated. enzymatic activity assayed, real-time quantitative polymerase chain reaction (RT-qPCR) performed to quantify levels. Results: In assays acid CoA ligase activity, caprylic found be optimal substrate for We determined catalytic conditions FadK, which include pH 7.4, ATP concentration 0.6 mM, 0.8 Mg2+ mM at 37 °C. Notably, showed decrease with increasing concentrations dodecyl-AMP, further confirmed RT-qPCR results. Conclusions: Our findings will fundamental framework development innovative therapeutics that target E. infections.

Drought stress significantly influences the physiological, biochemical, and molecular processes in plants, directly impacting their growth defense mechanisms. This study evaluates response of Lavandula angustifolia (lavender) to different levels water deficit, with field capacity (FC) treatments set at 20%, 40%, 60%, 80%. We assessed various biochemical parameters, including protein content, chlorophyll a b levels, flavonoid phenolic antioxidant activity, determine how drought affects lavender's primary secondary metabolism. As availability decreased, we observed reduction total while highest flavonoids, phenolics, activity were recorded control plants 80% FC. Gene expression analysis key terpene synthase genes revealed differential patterns, linalool α-pinene peaking 40% FC, 1,8-cineole β-phellandrene reaching under severe (20% FC). Despite this, clear correlation between gene metabolite accumulation essential oils was not observed. Drought-induced changes oil composition associated enhanced antibacterial particularly against foodborne Gram-positive Gram-negative bacteria, suggesting that can modulate therapeutic potential lavender oil.
