Abstract Salivary biomarkers can improve the efficacy, efficiency, and timeliness of oral maxillofacial disease diagnosis monitoring. Oral conditions in which salivary have been utilized for disease‐related outcomes include periodontal diseases, dental caries, cancer, temporomandibular joint dysfunction, gland diseases. However, given equivocal accuracy during validation, incorporating contemporary analytical techniques biomarker selection operationalization from abundant multi‐omics data available may help performance. Artificial intelligence represents one such advanced approach that optimize potential to diagnose manage Therefore, this review summarized role current application based on artificial discovery validation

Chronic psychological stress is known to influence immune function and contribute development of autoimmune disorders through dysregulated inflammatory responses. This study investigates relationships between acute stress, life trauma, salivary biomarkers in firefighter recruits during psychophysical training. Salivary samples were collected from two tests evaluate responses stress. Samples obtained at three time points—pre-stress, post-stress, recovery—across both tests. Cortisol was measured characterize response (ASR) profiles, while assessed the analyzing C-reactive Protein (CRP), Complement C4 (C4), Pigment Epithelium Derived Factor (PEDF), Serum Amyloid P (SAP). Results showed significant changes CRP, C4, PEDF after inoculation. Higher previous trauma associated with lower baseline cortisol (r = −0.489) delay recovery 0.514), suggesting a learned biological response, potentially protective against stress-induced dysregulation. Cluster analysis revealed four distinct ASR profiles which found have significantly different past (p 0.031). These findings suggest that history influences biomarker dynamics, reflecting individualized adaptive or maladaptive The insights gained may inform strategies enhance resilience mitigate risk among high-stress populations.

Intervertebral disc degeneration (IVDD) is one of the most prevalent causes chronic low back pain. The role m6A methylation modification in remains unclear. We investigated immune-related regulators as IVDD biomarkers through comprehensive analysis and experimental validation degeneration. training dataset was downloaded from GEO database analysed for differentially expressed immunological features, were subsequently validated by a rat model RT-qPCR. Further screening key based on machine learning LASSO regression analysis. Thereafter, predictive constructed sets, which set. patients then clustered expression regulators, immune infiltrates between clusters to determine markers IVDD. Finally, we potential marker enrichment analysis, protein-to-protein network molecular prediction. By analysising set, revealed significant differences gene five including RBM15, YTHDC1, YTHDF3, HNRNPA2B1 ALKBH5, while finding characteristic infiltration genes, result PCR. screen differential set identified RBM15 YTHDC1 regulators. used construct successfully it Next, explored characteristics well clusters. finally an biomarker found that may influence microenvironment ABL1 TXK. Multiple studies have demonstrated that human neurobiology and behavior are inextricably linked to the activity of our immune systems. Trauma is associated with a multitude system changes; reflecting this, posttraumatic stress disorder (PTSD) often comorbid immune-related conditions such as autoimmune disorders. To further investigate this phenomenon, we tested hypothesis cytokine fluctuations during after an acute response correlates experienced life trauma.Using prospective observational approach, cohort study measured biomarker profiles in firefighter participants (n = 63), 9 having prior PTSD diagnoses 54 without diagnoses. In addition, trauma scores were determined from all using Life Events Checklist 5 (LEC-5) survey. Baseline salivary concentrations determined, along levels immediately before, after, 1 hour following standardized stressful training event. Biomarkers these samples included 42 cytokines 6 steroid thyroid hormones. The markers then correlated, Pearson correlation coefficients, participants' LEC-5 scores. t Tests also performed compare values between populations diagnosis.Included panel interleukin (IL)-8, IL-10, IL-1B, GCSF, IL1-Ra, Groα, IFNa2, PDGFAA, VEGF, which positive at various time points individuals increased severity (and thus trauma). Concentrations IL-1a, Mip1a, IL-6, Mip1b, TNFα, TGFα found be significantly altered diagnoses, demonstrating some overlap correlations. The results support demonstrate indeed correlated surrounding test.Diagnostic or Criteria; Level IV.

Frequent exposure to acute stress increases risk of suicide, posttraumatic disorder, and other stress-related disorders. Neuroendocrine immunologic dysregulation associated with may underlie predispositions psychological disorders inflammatory disease processes in individuals, such as first-responders healthcare professionals, who function high situations. The Hardiness Resilience Gauge (HRG) can be used psychometrically measure resilience, a modifier the response. Using HRG alongside salivary biomarker profiling, help identify low resilience phenotypes allow mitigation early therapeutic interventions. There is paucity knowledge regarding biomarkers resilience. This study aims evaluate relationship between factors levels fluctuations during following stress.Sixty-three first responders underwent standardized stress-inducing training exercise, providing samples before (prestress), immediately after (post-stress), 1 hour event (recovery). was administered (initial) (final) event. Multiplex ELISA panels quantified 42 cytokines 6 hormones from samples, which were analyzed for relationships psychometric measured by HRG.Several correlated scores ( p < 0.05) select set moderate-to-strong correlations (|r| > 0.3). These included EGF, GROα, PDGFAA, TGFα, VEGFA, interleukin (IL)1Ra, TNFα, IL18, cortisol, FGF2, IL13, IL15, IL6. Interestingly, PDGFAA post-stress compared recovery positively negatively pre-stress period. This exploratory analysis discovered small subset that are significantly Further investigation their specific roles associations resiliency warranted.

Once thought of as an immune-privileged site, we now know that the nervous system communicates in a bidirectional manner with immune via neuroimmune axis. Neuropeptides constitute component this axis, playing critical roles brain and periphery. The function salivary neuropeptides acute stress response is not well understood. purpose study to investigate neuropeptide levels during stress. Salivary samples were collected from fire recruits engaged training exercise previously shown induce stress, at three separate timepoints oxytocin, neurotensin, Substance P, α-MSH, β-Endorphin measured using Human Neuropeptide 5-Plex Custom Assay Eve Technologies. All increased throughout simulation recovery phase. Exploratory factor analysis (EFA) identified one contributing baseline values across five Pairwise Pearson Correlation Coefficient showed positive correlations >0.9 for almost all combinations pre-stress timepoint. Further negative between past-life trauma self-assessed hardiness, respectively. Calculated scores overall correlation hardiness. Altogether, our results suggest increase synchronously higher correlate required determine if interventions designed enhance activity can resilience, especially high-stress occupations such firefighting.

Objective: The objective of this study was to investigate possible immune cytokine trends throughout a week-long surgical simulation mass-casualty training session in order determine the effects stress inoculation on system. Methods: Thirty-seven military medical students participated hyper-realistic event conducted at Strategic Operations site San Diego, California. Salivary samples were collected every morning exercise for 4 consecutive days. Cortisol, along with panel 42 cytokines, measured using multiplex enzyme-linked immunosorbent assays from Eve Technologies. determined concentrations averaged and plotted scatter plot, then points fit second-order polynomial trendline best measure. Results: cytokines epidermal growth factor, growth-related oncogene-α, interleukin (IL)-1α, platelet-derived factor-AA followed noted pattern cortisol decrease week. In addition, IL-27, granulocyte colony stimulating IL-10, IL-13 demonstrated late peak, by return baseline conclusion training. Finally, monocyte chemoattractant protein-1 displayed decline week an increase last day training. Conclusions: Altogether, these results help identify important biomarkers that may improve long-term adaptation prevent post-traumatic disorder following exposure repeated stress.
