Rapid
vaccine
development
and
innovative
immunotherapeutics
are
critical
in
the
fight
against
emerging
outbreaks
global
pandemic
threats,
yet
high
costs
prolonged
timelines
for
developing
new
vaccines
underscore
urgent
need
robust,
predictive
pre-clinical
testing
platforms.
The
rapid
down-selection
of
candidates
identification
optimal
formulations
can
be
performed
using
human
vitro
immunization
(IVI)
assays
that
recapitulate
complex
interactions
innate
adaptive
immune
response.
In
this
review,
we
present
a
comprehensive
evaluation
three
key
IVI
platforms:
whole
blood
assay
(WBA),
monocyte-derived
dendritic
cell
(MoDC)
with
cell-T
interface
(DTI),
microphysiological
tissue
construct
(HTC).
WBA
offers
cost-effective
straightforward
approach,
while
MoDC
+
DTI
system
represents
current
gold
standard
balancing
experimental
efficiency
immunological
complexity.
HTC
assay,
by
mimicking
both
spatial
temporal
aspects
interactions,
provides
enhanced
physiological
relevance.
We
discuss
methodological
advantages
limitations
each
platform,
explore
their
roles
candidate
screening,
propose
strategies
integrating
these
complementary
vivo
models.
These
insights
pave
way
refining
accelerating
translational
pipeline
next-generation
immunotherapies.
