Investigating human pathogens in wastewater is crucial for identifying and predicting potential occupational health risks faced by treatment plant (WWTP) workers. This study aimed to determine the occurrence levels of Legionella pneumophila, Mycobacterium spp., Arcobacter butzleri, Aeromonas hydrophila untreated municipal wastewater. Grab influent, activated sludge, secondary settling tank (SST) effluent samples were collected bi-weekly over 6 months from 5 WWTPs Tshwane, South Africa. Geographical variations in infectious diseases create differences public health priorities between high- and low-income countries. Low- middle-income countries (LMICs) face resource constraints that limit adherence to international monitoring standards for wastewater-based epidemiology (WBE). Abstract The COVID-19 pandemic has underscored the importance of virus surveillance in public health and wastewater-based epidemiology (WBE) emerged as a non-invasive, cost-effective method for monitoring SARS-CoV-2 its variants at community level. Unfortunately, current variant methods depend heavily on updated genomic databases with data derived from clinical samples, which can become less sensitive representative testing sequencing efforts decline. Laboratory testing has played a pivotal role throughout the coronavirus disease 2019 (COVID-19) pandemic, exemplifying importance of in vitro diagnostics addressing public health threats posed by outbreaks infectious diseases. This article aims to present key insights from our expertise, derived evidence gathered during COVID-19 inform strategies for managing future challenges. Current scientific underscores that patient sample not only allows diagnose an acute severe respiratory syndrome 2 (SARS-CoV-2) infection, but also supports outbreak prediction, improved control measures, anticipation pressure on healthcare system, mitigation adverse clinical outcomes, and early detection emerging SARS-CoV-2 variants. Additionally, wastewater monitoring emerged as powerful tool forecasting burden, including both prevalence severity. Collectively, these findings underscore value diagnostic surveillance guiding planning optimizing resource allocation offering valid framework be applied threats, especially any potential “Disease X” may emerge future.

The increasing use of genetic testing for personalised therapy, highlights the need rapid, reliable diagnostics. Current methods are hindered by complex workflows, requiring advanced equipment, skilled personnel, and invasive tissue sampling. Loop-mediated isothermal amplification (LAMP) has emerged as a more efficient alternative to traditional PCR. LAMP eliminates thermal cycling, allowing faster, cost-effective tests, is less sensitive inhibitors, enabling from minimally processed samples. Although newer longer assay development time than PCR, its potential in oncology, particularly detecting changes, promising. We have developed LAMP-based method variations, optimized point-of-care testing. This technique uses modified primers with alterations at 3' end either F2 or B2 primers, ensuring specificity altered sequences. only produces positive signal when variant present, distinguishing it wild-type DNA. Our findings demonstrate that this high sensitivity, even samples both mutated material. Paired portable device, diagnostic could revolutionize alteration detection, offering quicker results improving treatment outcomes, targeted therapies.
