The quest for eternal youth and immortality is as old humankind. Ageing an inevitable physiological process accompanied by many functional declines that are driving factors age-related diseases. Stem cell exhaustion one of the major hallmarks ageing. SOX transcription play well-known roles in self-renewal differentiation both embryonic adult stem cells. As a consequence ageing, repertoire cells present various organs steadily declines, their dysfunction/death could lead to reduced regenerative potential development Thus, restoring function aged cells, inducing potential, slowing down ageing critical improving health span and, consequently, lifespan humans. Reprograming factors, including family members, emerge crucial players rejuvenation. This review focuses on diseases, neurodegenerative visual deterioration, chronic obstructive pulmonary disease, osteoporosis, cancers. With geroscience research evolving at a fast pace, the need arises for human randomized controlled trials to assess efficacy of geroprotective interventions prevent age-related adverse outcomes, disease, and mortality in normative aging cohorts. However, confirm requires long-term costly approach as time event morbidity can be decades. While this could circumvented using sensitive biomarkers aging, current molecular, physiological, digital endpoints require further validation. In review, we discuss how collecting real-world evidence (RWE) by obtaining health data that is amenable collection from large heterogeneous populations setting help speed up validation interventions. Further, propose inclusion quality life (QoL) biomarker candidate endpoint clinical aid distinguishing healthy unhealthy aging. We highlight QoL assays accelerating studies gathering RWE on effects repurposed drugs support utilization within longevity medicine. Finally, summarize key metrics consider when implementing studies, present short-form 36 (SF-36) most well-suited endpoint.

Studying aging now requires going beyond the bio-psycho-social model and incorporating a broader multidisciplinary view capable of capturing ultimate complexity being human that is expressed as individuals age. Current demographic trends lengthening life expectancies allow observation long-lived in full health. These super-agers are no longer an exception. Indeed, can have good quality even over age 70 living with chronic or neurodegenerative diseases. This change driven part by cohort effect observed people who about to today (e.g., better schooling, more advanced health conditions, technologization) but so gradual overcoming ageist views. An aged person seen quitter rather one empowered direct their own trajectory potentially healthy longevity. According this vision, article proposes situated lifespan perspective for study integrates pedagogical models developmental ecology psychological theories optimal experience understand individual motivational on aging. At same time, it does not disregard analyzing daily cultural contexts which everyone situates guide trajectories. Aging involves tissue and cell potential dysfunction characterized by stem senescence extracellular matrix microenvironment (ECM) alteration. Chondroitin sulfate (CS), found in the ECM of normal cells tissues, aids maintaining homeostasis. Here, CS-derived biomaterial (CSDB) from sturgeon is extracted to investigate its antiaging effect senescence-accelerated mouse prone-8 (SAMP8) mice elucidate underlying mechanism action. Although CSDB has been widely different sources used as a scaffold, hydrogel, or drug carrier for treatment various pathological diseases, not yet amelioration aging features. In this study, showed low molecular weight comprised 59% 4-sulfated CS 23% 6-sulfated CS. an vitro promoted proliferation reduced oxidative stress inhibit senescence. ex vivo after oral SAMP8 mice, were analyze p16Ink4a p19Arf gene-related pathways, which inhibited then SIRT-1 gene expression was upregulated reprogram state retarding aging. also restored aging-phenotype-related bone mineral density skin morphology prolong longevity. Thus, may be useful prolonging healthy longevity anti-aging drug.

Aging, an inevitable physiological process leading to a progressive decline in bodily functions, has been abundantly researched domain with studies attempting slow it down and reduce its debilitating effects. Investigations into the cellular molecular pathways associated aging have allowed formulation of therapeutic strategies. Of these, caloric restriction (CR) implicated for role promoting healthy by modulating key targets like Insulin/IGF-1, mTOR, sirtuins. However, CR requires dedication commitment strict regimen which poses difficulty maintaining consistency. To maneuver around cumbersome diets, Caloric Restriction Mimetics (CRMs) emerged as promising alternatives mimicking beneficial effects CR. This review elucidates foundations enabling CRMs rapamycin, metformin, resveratrol, spermidine, many more function suitable anti-aging molecules. Moreover, explores clinical trials (retrieved from clinicaltrials.gov database) aimed at demonstrating efficacy effective candidates against age-related neurodegenerative diseases such Alzheimer's disease Parkinson's disease.

Decreased content and activity of the mechanistic target rapamycin (mTOR) signalling pathway, as well mTOR complex 1 (mTORC1) itself, are key traits for animal species human longevity. Since mTORC1 acts a master regulator intracellular metabolism, it is responsible, at least in part, longevous phenotype. Conversely, increased hallmarks ageing. Additionally, constitutive aberrant also found age-related diseases such Alzheimer’s disease (AD) cancer. The downstream processes regulated through this network diverse, depend upon nutrient availability. Hence, multiple nutritional strategies capable regulating and, consequently, delaying ageing process development diseases, under continuous study. Among these, restriction calories still most studied robust intervention downregulating feasible application population.

Beyond exhaustion pension funds budgets, by reviewing literature analysing publicly available data, we observe "Swiss Japanification". Old age is late-life comorbidities increasing proportion time spent poor health. To address these problems, paradigm practice needed improve health rather than respond existing diseases. One major player of adaptive response are T cells, recent studies argue for role disturbed proteostasis contributing reduced function these cells upon aging. Proteostasis refers state healthy, balanced proteome is influenced by synthesis (translation), maintenance quality control proteins, as well degradation damaged or unwanted proteins proteasome, autophagy, lysosome cytoplasmic enzymes. This review focuses on molecular processes impacting specifically functional quantitative changes each Importantly, we describe biological consequences compromised range from impaired activation enhancement inflamm-aging aged cells.
