Attention deficit hyperactivity disorder (ADHD) is a common neurodevelopmental in children and adolescents, its etiology pathogenesis are still unclear. This study probed the ameliorative effects of naringenin in a D. melanogaster model 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced parkinsonism, incorporating computational analysis. Initially, flies were treated with (100-500 µM) and MPTP (250-750 for 14 days two separate studies to determine optimum concentrations treatments. Following this, (100 300 administered (500 µM)-exposed 4-day study. Motor function, survival rate, neurotoxicity biomarkers assessed alongside biological network analysis molecular docking simulation. Results indicate that exhibits hormetic behavior, 100-300 µM providing optimal neuroprotection. The treatments significantly improved negative geotaxis acetylcholinesterase activity, reduced MPTP-induced oxidative stress as indicated by nitric oxide, hydrogen peroxide, protein carbonyl levels. Furthermore, restored thiol contents, enhanced catalase glutathione-S-transferase activities. Network helped identify key targets, including DRD4, DRD2, NFKB1, MAOB, MAPK14, CYP2A6, which function dopaminergic signaling oxido-inflammatory pathways. Molecular revealed strong binding interactions MAO, MAPK, NF-κB primarily through bonding hydrophobic interactions. Overall, these findings suggest mitigates enhancing neurotransmission suppressing inflammation. further supports neuroprotective potential could be suggested promising nutraceutical/drug candidate Parkinson's disease.

There reportedly exists a significant comorbidity between insomnia and neurodegenerative diseases, such as Alzheimer's disease (AD) Parkinson's (PD), indicative of potential link to serum metabolic dysregulation. To elucidate shared pathophysiological mechanisms AD/PD, we performed comprehensive two-sample Mendelian randomization (MR) analyses, investigating 1,400 characteristics for their causal relationships with the risks insomnia, AD, widely defined AD (WDAD), PD. We employed publicly available genetic data; primary estimate was determined using inverse-variance weighting, supplemented by weighted median, simple mode, MR-PRESSO MR-Egger methods evaluate heterogeneity pleiotropy. This study aimed to assess the association between motor and non-motor symptoms of Parkinson's disease (PD) iron accumulation within deep gray matter brain by Quantitative Susceptibility Mapping (QSM). Fifty-six PD patients twenty-nine healthy controls were recruited in this study. According Hoehn Yahr (H-Y) stage score, divided into early (H-Y ≤ 2) advanced > groups. Specifically, Regions Interest (ROIs) encompassed substantia nigra (SN), red nucleus (RN), caudate (CN), globus pallidus (GP) putamen (PT). Meanwhile, various rating scales used clinical PD. Compared (HCs), showed a significant increase magnetic susceptibility values (MSVs) SN GP. Further comparisons indicated that MSVs SN, PT, GP CN are all higher stages than stages. Significant positive correlations observed scores on UPDRS-III, HAMA, HAMD (r = 0.310, p 0.020; r 0.273, 0.042; 0.342, 0.010, respectively). Likewise, demonstrated notable with HAMA 0.275, 0.040; 0.415, 0.001). Additionally, correlation was found PT 0.360, 0.006). Furthermore, we identified negative MMSE both -0.268, 0.046; -0.305, 0.022). Our revealed QSM possesses capability serve as biomarker for features deposition nigrostriatal system.

ABSTRACT Artificial light sources, particularly blue light, have raised concerns about their impact on biological health and behavior. In this study, we explored the effects of locomotion cognitive functions early adult Drosophila melanogaster . Our experiments were conducted in a custom‐designed behavioral arena to assess how influences these parameters. We found that after ~2 weeks exposure, fruit flies, including movement speed activity levels, significantly increased, but disappeared by third week. However, exposure also led notable reduction lifespan had adverse functions, attention short‐term spatial memory. These findings may reveal profound impacts neural lifespan, highlighting importance further research into light.

Cerebral vascular disorders often accompany hypoxia-induced brain injury. In this study, we develop a zebrafish model of cerebral injury to replicate the associated phenotypic changes, including cerebrovascular damage, neuronal apoptosis, and neurological dysfunction. We then explored therapeutic potential extracellular vesicles derived from Wharton's jelly-derived mesenchymal stem cells (WJ-MSCs) cultured on soy protein-coated surfaces. These demonstrated superior recovery efficacy, especially in restoring blood–brain barrier integrity improving function. Our findings suggest that these potent vesicles, easily produced WJ-MSCs presence proteins, may mitigate by decreasing severity disorder caused oxidative stress. Protein–protein interactome analysis further suggests multiple signaling pathways are likely involved normal neurovascular unit

Oxidative stress and neuroinflammation are considered as the two main etiological reasons behind idiopathic Parkinson’s disease (PD). Nevertheless, actual treatments focused on improving motor symptoms by restoring dopamine (DA) presence, leaving said causes unattended. Probiotics could be a promising strategy for improvement of these physiological features therefore constitute complementary treatment those having PD. This study evaluated effect oral administration probiotic bacteria mixture from 3 strains Limosilactobacillus fermentum LH01 , reuteri LH03, Lactiplantibacillus plantarum LH05 (LLH135), human milk origin, 4 weeks, mice under hemiparkinsonism model intrastriatal 6‐hidroxidopamine (6‐OHDA). We measured total antioxidant capacity (TAC), super oxide dismutase (SOD) activity, 8‐deoxyguanosine (8‐OHdG) regarding oxidative stress. Concerning neuroinflammation, immunoreactivity GFAP, IBA‐1, CD68 was immunohistochemistry latter markers corroborated in colocalization with immunofluorescence to assess activated microglia. The diminished SOD activity well 8‐OHdG generated hemiparkinsonism. These effects were accompanied dampening glial IBA‐1 that present model. Our findings suggest LLH135 exerts neuroprotective promoting an response which explained modulation cells dopaminergic neuronal damage induced 6‐OHDA.
