Chimeric Antigen Receptor (CAR) T-cell therapy has revolutionized the treatment of hematological malignancies, demonstrating high efficacy in targeting and eliminating cancer cells. However, its clinical application can be associated with risk acute adverse effects, including cytokine release syndrome (CRS), a severe inflammatory response caused by excessive production. While anti-cytokine therapies are available to manage CRS, additional strategies needed optimize CAR reduced toxicities. Curcumin, bioactive polyphenol known for anti-inflammatory antioxidant properties, represents promising adjunct therapy. In this study, we investigated effects curcumin on anti-CD19 T-cells vitro. Our results show that enhances cytotoxic activity against Nalm-6, B-cell lymphoblastic leukemia model, while reducing production pro-inflammatory cytokines, IL-2 IFN-γ. To explore underlying mechanisms, network pharmacology molecular docking analyses were performed, which revealed interacts key signaling pathways involved activation regulation. These findings support potential as therapeutic improve mitigating toxicity.

Systemic chemotherapy is still the first-line treatment for cancer, and it’s associated with toxic side effects, chemoresistance, ultimately cancer recurrence. Rapid gelling hydrogels can overcome this limitation by providing localized delivery of anti-cancer agents to solid tumors. Silk are extremely biocompatible suitable drug delivery, but faster formulations needed. In study, we introduce a rapid hydrogel formulation (<3 min time) due chemical crosslinking between silk fibroin curcumin, initiated addition minute quantities horseradish peroxidase (HRP) hydrogen peroxide (H2O2). The novel observation in study that while being free-radical scavenger, also participates accelerating di-tyrosine presence HRP H2O2.
