Abstract Growth factor receptor‐bound protein 2 (GRB2) is a key adaptor involved in multiple signalling pathways, and its dysregulation associated with various diseases. Type diabetes systemic condition characterized by insulin resistance impaired β‐cell function. The complications of significantly reduce life expectancy quality life, imposing substantial burden on society. However, the role GRB2 largely unknown. Emerging evidence suggests that plays crucial resistance, inflammation, immune activation regulation cellular processes such as cell proliferation, growth, metabolism, angiogenesis, apoptosis differentiation. Dysregulation GRB2‐mediated pathways contributes to progression diabetic neuropathy, cognitive dysfunction, nephropathy, retinopathy related disorders. This review provides comprehensive overview current understanding diabetes, disorders, alongside recent advances development GRB2‐targeted therapies. Elucidating complex these disorders valuable insights into potential therapeutic strategies targeting pathways.
