Chronic kidney disease (CKD) progresses through mechanisms involving inflammation, fibrosis, and oxidative stress, leading to the gradual structural functional deterioration of kidneys. Tormentic acid (TA), a triterpenoid compound with known anti-inflammatory antioxidant properties, shows significant potential in counteracting these pathological processes. This study explored protective role TA unilateral ureteral obstruction (UUO)-induced CKD model. Mice received intraperitoneal injections at dosage 5 mg/kg per day for 8 consecutive days, commencing before UUO procedure. The treatment significantly improved both injury. It suppressed cytokine expression reduced immune cell infiltration, inhibited activation mitogen-activated protein kinase cascade, alleviated endoplasmic reticulum stress. Moreover, displayed potent anti-fibrotic effects by reversing epithelial-to-mesenchymal transition inhibiting Smad2/3 activation, reducing extracellular matrix deposition. also mitigated stress attenuating lipid peroxidation boosting defenses. Additionally, it apoptosis ferroptosis modulating key death markers. Collectively, findings indicate that provides comprehensive renoprotection model effectively targeting tubular progression.

The purpose of the study was to investigate renal oxidative stress (OS) and its relationship with disease severity in patients chronic kidney (CKD) using positron emission tomography coupled magnetic resonance imaging (PET/MRI), employing 64Cu-diacetyl-bis(N4-methylthiosemicarbazonate) (64Cu-ATSM) as PET tracer for OS imaging. Thirty CKD (66.4 ± 8.2 y.o.) seven healthy controls (HC) subjects (58.3 3.8 underwent 64Cu-ATSM PET/MRI. Participants were categorized into three groups based on their estimated glomerular filtration rate (eGFR): HC, mild (stages 2-3a), advanced 3b-5). All 30-min dynamic PET/MRI starting injection evaluate blood flow (RBF) levels. RBF (mL/min/100 g) images calculated from first 3 min data, standardized uptake value (SUV) obtained delayed frames 15-30 after injection. SUV corrected RBF-adjusted individual estimate levels kidneys following equation: adjusted index (aOSi) = (SUV/RBF)x100. Significant correlation observed between eGFR (r 0.81, P < 0.001). is significantly lower than that HC (P 0.001) 0.004). did not differ among 0.171). SUVs correlate creatinine or CKD. However, these values 0.33, 0.049) all subjects, whereas showed no significant correlation. Following correction, aOSi demonstrated correlations 0.75, 0.001), (r= -0.65, stages 0.57, subjects. This preliminary has revealed may provide a reasonably noninvasively. Increased correlated levels, suggesting increases dysfunction.

In 2008, the Society on Sarcopenia, Cachexia, and Wasting Disorders introduced a generic definition for all types of cachexia: "a complex metabolic syndrome associated with underlying illness characterized by loss muscle, or without fat loss". It is well-known that presence inflammatory burden in end-stage renal disease (ESRD) patients may lead to evolution cachexia. Since etiology cachexia chronic kidney (CKD) multifactorial, thus successful treatment must involve several concomitant measures (nutritional interventions, appetite stimulants, anti-inflammatory pharmacologic agents) provide integrated effective therapeutic modalities combat causative factors alleviate outcomes patients. Given high mortality rate cachexia, developing new are prerequisite ameliorating CKD worldwide. The present review aims discuss some strategies an update advances nutritional approaches counteract

Abstract Objective This study aims to investigate the association between docosanoic acid (DA) in human circulation and risk of occurrence mortality chronic kidney disease (CKD). Methods is a cross-sectional including individuals NHANES 2011-2014. RCS were used fit dose-effect curve DA levels CKD general population, patients. Multiple logistic Cox regressions analyze with risks respectively. Results A total 2,366 participants included this study, 1,958 (82.8%) without 408 (17.2%) individuals. The results showed linear Per standard deviation (per-SD) increase DA, population decreased by 18% (OR = 0.82; 95% CI, 0.70 - 0.96; P 0.02). In addition, cardiovascular all-cause CKD. Multivariate regression analysis that per-SD patients 44% (HR 0.56; 0.35 0.90; 0.02) 27% 0.73; 0.59 0.89; 0.002). Conclusion Higher serum populations means lower risk. Moreover, for patients, as increase, decreases.

Chronic kidney disease (CKD) is a significant public health concern. Osteoarthritis (OA), common form of arthritis, has been shown to have dramatically increased prevalence, particularly among individuals aged 40-50 and older, in the presence CKD. Furthermore, CKD may exacerbate progression impact OA. A survey study revealed that 53.9% patients undergoing long-term hemodialysis were diagnosed with These findings underscore potential association between Uremic toxins, such as indoxyl sulfate, p-cresyl transforming growth factor-β, advanced glycation end-products, are regarded risk factors various CKD-related conditions, affecting bone joint metabolism. However, whether these serve bridging mechanism OA comorbidities, well their detailed roles this context, remains unclear. Addressing identifying effective treatment prevention strategies an urgent challenge warrants immediate attention. This review focuses on describing discussing molecular pathological mechanisms underlying CKD-associated possible therapeutic strategies.

Purpose: Vaccarin is a natural flavonoid glycoside with anti-inflammatory, antioxidant and nephroprotective effects. However, the effects of vaccarin on renal fibrosis (RF) its molecular mechanisms remain unclear. This study aimed to investigate RF mechanisms. Methods: Network pharmacology was used analyze effect RF, docking dynamics simulations were performed assess binding nuclear factor erythroid 2-related 2 (Nrf2) vaccarin. A mouse model unilateral ureteral obstruction (UUO) established in vivo, human tubular epithelial (HK2) cells induced transforming growth factor-β (TGF-β) RSL3, respectively, as an vitro model. The anti-fibrotic observed by histopathological staining determination fibrous markers. Changes oxidative stress ferroptosis-related markers detected kits, Western blot (WB), qRT-PCR immunofluorescence (IF). Finally, Nrf2 inhibitors added observe ferroptosis. Results: cross genes are enriched for stress. binds stably Both vivo experiments showed that treatment reduced expression markers, decreased levels reactive oxygen species (ROS), malondialdehyde (MDA), lipid peroxidation (LPO) Fe 2+ , increased glutathione (GSH) secretion. In addition, down-regulated Long-chain acyl-CoA synthetase 4 (ACSL4), prostaglandin-endoperoxide synthase (PTGS2) NADPH oxidase 1 (NOX1), up-regulated downstream solute transport family 7 member 11 (SLC7A11) peroxidase (GPX4) expression. Mechanistic studies indicated activated Nrf2/SLC7A11/GPX4 pathway inhibit ferroptosis, this inhibition effectively reversed inhibitor. Conclusion: ameliorates inhibiting ferroptosis via pathway.
