Chronic kidney disease (CKD) progresses through mechanisms involving inflammation, fibrosis, and oxidative stress, leading to the gradual structural functional deterioration of kidneys. Tormentic acid (TA), a triterpenoid compound with known anti-inflammatory antioxidant properties, shows significant potential in counteracting these pathological processes. This study explored protective role TA unilateral ureteral obstruction (UUO)-induced CKD model. Mice received intraperitoneal injections at dosage 5 mg/kg per day for 8 consecutive days, commencing before UUO procedure. The treatment significantly improved both injury. It suppressed cytokine expression reduced immune cell infiltration, inhibited activation mitogen-activated protein kinase cascade, alleviated endoplasmic reticulum stress. Moreover, displayed potent anti-fibrotic effects by reversing epithelial-to-mesenchymal transition inhibiting Smad2/3 activation, reducing extracellular matrix deposition. also mitigated stress attenuating lipid peroxidation boosting defenses. Additionally, it apoptosis ferroptosis modulating key death markers. Collectively, findings indicate that provides comprehensive renoprotection model effectively targeting tubular progression.
