Preeclampsia (PE) is a complex pregnancy syndrome characterized by hypertension with or without proteinuria, affecting 2–6% of pregnancies globally. Preeclampsia, a serious and potentially life-threatening medical complication occurring during pregnancy, is characterized by hypertension often accompanied proteinuria multiorgan dysfunction. It classified into two subtypes based on the timing of diagnosis: early-onset (EO-PE) late-onset preeclampsia (LO-PE). Despite being less severe exhibiting distinct pathophysiological characteristics, LO-PE more prevalent than EO-PE, although both conditions have significant impact placental health. Previous research indicates that different events within placenta may contribute to development across multiple pathways. In our experimental study, we investigated markers oxidative stress, ferroptosis, lipid peroxidation pathways in tissue samples obtained from women with (n = 68) compared healthy control pregnant (HC, n 43). Through comprehensive analysis, observed an upregulation specific molecules associated these pathways, including NADPH oxidase 1 (NOX-1), 2 (NOX-2), transferrin receptor protein (TFRC), arachidonate 5-lipoxygenase (ALOX-5), acyl-CoA synthetase long-chain family member 4 (ACSL-4), glutathione peroxidase (GPX4) malondialdehyde (MDA) LO-PE. Furthermore, increased ferric deposition (Fe3+) was stained Perls’ Prussian blue. The assessment involved gene expression analyses conducted through RT-qPCR experiments immunohistochemistry assays. The nucleotide-binding domain, leucine-rich repeat, and pyrin domain containing-protein 3 (NLRP3) inflammasome is a multiprotein complex that plays critical role in the innate immune response to both infections sterile stressors. Dysregulated NLRP3 activation has been implicated variety of autoimmune inflammatory diseases, including cryopyrin-associated periodic fever syndromes, diabetes, atherosclerosis, Alzheimer’s disease, bowel cancer. Consequently, fine-tuning activity holds significant therapeutic potential. Studies have several organelles, mitochondria, lysosomes, endoplasmic reticulum (ER), Golgi apparatus, endosomes, centrosome, localization assembly. However, reports conflict many factors regulating interactions between subcellular organelles remain unknown. This review synthesizes current understanding spatiotemporal dynamics, focusing on recent literature elucidates roles organelle stress signaling its crosstalk with other pathways converging at these organelles.

Chronic venous disease (CVD) comprises a spectrum of morphofunctional disorders affecting the system, approximately 1 in 3 women during gestation. Emerging evidence highlights diverse maternofetal implications stemming from CVD, particularly impacting placenta. While systemic inflammation has been associated with pregnancy-related preliminary findings suggest potential link between this condition and exacerbated placental tissue. Inflammasomes are major orchestrators immune responses different organs systems. Notwithstanding relevance inflammasomes, specifically NLRP3 (nucleotide-binding domain, leucine-rich-containing family, pyrin domain-containing-3)- which demonstrated placentas obstetric complications, precise involvement component CVD remains to be explored. Preeclampsia (PE) is a complex multisystem disease characterized by hypertension of sudden onset (>20 weeks' gestation) coupled with the presence at least one additional complication, such as proteinuria, maternal organ dysfunction, or uteroplacental dysfunction. Hypertensive states during pregnancy carry life-threatening risks for both mother and baby. The pathogenesis PE develops due to dysfunctional placenta aberrant architecture that releases factors contributing endothelial an antiangiogenic state, increased oxidative stress, inflammatory responses. Previous studies have shown correlation between grade 3 placental calcifications elevated risk developing term. However, little known about molecular pathways leading calcification.
