Three-dimensional bioprinting allows for the fabrication of structures mimicking tissue architecture. This study aimed to develop a gelatin-based bioink bioprinted simplified skin model. The printability and chemical-physical properties were evaluated by varying concentrations gelatin (10, 15, 20%) in semi-crosslinked alginate-based calcium chloride (100, 150, 200 mM) post-printing crosslinking. For increasing concentration, formulations have shear thinning behavior with viscosity, filament bending angle increases, spreading ratio value approaches 1, shape fidelity printing resolution improve. Diabetic foot ulcers (DFU) affect up to 15-25% of patients suffering from diabetes and are considered a global health concern. Background/Objectives: There is a great demand for novel, multipurpose, natural skin-care products in the global skin repair and sun protection markets. Within this framework, potential benefits of topical Vitamin D2 (VD2) administration combination with silver nanoparticles (AgNPs) were examined. Methods: Evaluating efficacy VD2+AgNP cream wound healing, irritation UVB irradiation necessitated preclinical testing using reconstructed human equivalent models (prepared from foreskins) containing both fully stratified epidermal layer underlying dermis. Results: Application significantly improved healing by stimulating keratinocyte re-epithelialization dermal fibroblast migration subjected to full-thickness (scratch biopsy punch) wounds, compared untreated models. The cream, administered prior induction 5% sodium dodecyl sulfate (SDS) afforded ameliorating cell viability thickness interleukin-1alpha levels. exposure (50 mJ/cm2) reduced (associated increased breakage), as well basal Ki67 supra-basal involucrin expression, CTRL sham-irradiated (protective capacity) showed greater minimizing damage.
