Alzheimer's disease (AD) is well known among the elderly and has a profound impact on both patients their families. Increasing research indicates that AD systemic disease, with strong connection to cardiovascular disease. They share common genetic factors, such as mutations in presenilin (PS1 PS2) apolipoprotein E (APOE) genes. Cardiovascular conditions can lead reduced cerebral blood flow increased oxidative stress. These factors contribute accumulation of Aβ plaques formation abnormal tau protein tangles, which are key pathological features AD. Additionally, deposits responses have been observed cardiomyocytes peripheral tissues. Abstract Background: Human immunodeficiency virus (HIV) infection is associated with significant metabolic disruptions, and understanding how these disruptions contribute to cardiovascular disease (CVD) essential for improving patient management therapeutic strategies. By integrating metabolomics transcriptomics, we aimed elucidate the cardiometabolic spectrum of people living HIV (PLWH) identify biomarkers pathways progression in this population. Methods: We conducted a cross-sectional observational study between October 2022 June 2023 at Third People’s Hospital Shenzhen, China. Participants were categorized into one four groups: HIV+ without abnormalities (healthy control [HC]), + untreated (UM) (HIV UM), treated (TM) TM), CVD CVD). Key transcriptomic features identified through comparative analysis using untargeted RNA sequencing, followed by statistical pathway enrichment analyses. ABSTRACT Major depressive disorder (MDD) involves complex neuroimmune interactions linked to gene modulation. Our study investigates synaptic-related dysregulation in peripheral blood mononuclear cells (PBMCs) from MDD patients, showing how these immune mirror neural processes. Using RNA-seq data, we identified 1,383 differentially expressed genes (DEGs) related crosstalk, with 49 DEGs effectively distinguishing patients controls based on synaptic functions. Synaptic genes, enriched for roles like vesicle transport, suggest mechanistic links between and signaling. Eleven were shared PBMCs brain regions involved mood regulation, highlighting a common molecular signature. Among them, ADORA3 RPS28 emerged as potential biomarkers.
