Bladder cancer (BC) occurrence and progression are accompanied by alterations in microRNAs (miRNAs) expression levels. Simultaneous detection of multiple miRNAs contributes to the accuracy reliability BC diagnosis. In this work, wrinkled silica nanoparticles (WSNs) were applied as microreactor for multiplex analysis without enzymes or nucleic acid amplification. Conjugated on surface WSNs, S9.6 antibody was adopted universal module binding DNA/miRNA duplexes, regardless their sequence. Furthermore, single-stranded DNA (ssDNA) labeled with quantum dots (QDs) identifying a given miRNA form QDs-ssDNA/miRNA, which enabled specific capture corresponding QDs WSNs. Based fluorescence signals that ultimately focused target could be sensitively identified femtomolar level (5 fM) wide dynamic range up 6 orders magnitude. The proposed strategy achieved high specificity obviously distinguish single-base mutation sequences possessed assay capability. Moreover, exhibited excellent practicability clinical serum specimens.

In this work, chitosan beads were used as a cost-effective platform for the covalent immobilization of unmodified single-stranded DNA, using glutaraldehyde cross-linking agent.
